目的 探讨依达拉奉抗谷氨酸诱导神经干细胞凋亡作用及发生机制.方法 采用10 μmol/L,浓度的谷氨酸作用于大鼠13.5 d胚胎皮层来源的神经干细胞,建立损伤模型.在建模同时采用依达拉奉进行干预,采用PI染色、Caspase-3染色及流式细胞仪检测.结果 在依达拉奉干预下,依达拉奉治疗组凋亡细胞阳性率、Caspase-3阳性率等凋亡相关指标均明显降低,与谷氨酸组相比有统计学意义.结论 依达拉奉可以拮抗谷氨酸诱导神经干细胞凋亡的效应,可能减低caspase-3,减少凋亡发生表达.%To investigate the effect of edaravone of neural stem cells (NSCs) against glutamate.Methods NSCs were cultured from E 13.5D SD rat embryo. This experiment was subjected to 3 groups: control group, glutamate group with 10 nmol/L glutamate for culturing 24 hs, and edaravone group with 10 umol/L glutamate and edaravone 500 ng/ml together for culturing 24 hs. PI staining, caspase-3 staining and flow cytometry were used to analyze. Results Compared with those in glutamate group, the positive apoptosis rate and positive caspase-3 rate significanfiy decreased (P<0.05). Conclusion Edaravone can reduce apoptosis of NSCs following glutamate through reducing the expression of caspase-3.
