竹节参皂苷对D-半乳糖致SH-SY5Y神经细胞损伤的保护作用研究

         

摘要

目的 研究竹节参皂苷保护D-半乳糖致SH-SY5Y神经细胞损伤的作用及机制.方法 采用50 g/L的D-半乳糖刺激SH-SY5Y神经细胞,以不同浓度竹节参皂苷(1,5,25 μg/mL)和50 g/L的D-半乳糖共同孵育SH-SY5Y神经细胞24 h.JC-1试剂盒检测细胞线粒体膜电位,Western blot检测细胞Sirt1蛋白的表达.结果 竹节参皂苷能显著升高线粒体膜电位和Sirt1蛋白的表达.结论 竹节参皂苷通过激活Sirt1抑制线粒体功能障碍从而改善D-半乳糖对SH-SY5Y神经细胞造成的损伤.%Objective To observe the protective effect and mechanism of Panax japonicus saponins on D-galactose induced SH-SY5Y neuron injury.Methods The 50 g/L D-galactose was used to stimulate SH-SY5Y neuron,Different concentrations Panax japonicus saponins (1,5,25 μg/mL) and D-galactose co-incubated with SH-SY5Y neuron for 24 h.The changes of mitochondrial membrane potential were identified by JC-1 detection.Protein level of silentmat ing type information regulation 2 Homolog-1 (Sirt1) was detected by Western blot analysis.Results Panax japonicus saponins significantly improved mitochondrial transmembrane potential and the expression of Sirt1 protein.Conclusion Panax japonicus saponins inhibits mitochondrial dysfunction by activating Sirt1,thereby improving D-galactose induced damage to SH-SY5Y neuron.
