PI3K信号通路中LCK、LAT、SYK、PKC基因在特发性眼眶炎性假瘤中差异表达分析

         

摘要

Objective To analyze the differential expression of LCK、LAT、SYK and PKC in lacrimal gland specimens of patients with idiopathic orbital inflammatory pseudotumor or simple orbital cavernous hemangioma, and provide reference for the pathogenesis of idiopathic orbital inflammatory pseudotumor. Methods Different expression of pathways on patients with Idiopathic Orbital Inflammatory Pseudotumor and orbital cavernous hemangioma were detected by gene chips. The expression of LCK、LAT、SYK、PKC in level of mRNA and protein were detected by PCR, immune histochemical exam and western blot in Idiopathic Orbital Inflammatory Pseudotumor and orbital cavernous hemangioma. Orbital cavernous hemangioma was considered as control group. Results Compared with control group, the experimental group was found that the expression of PI3K signaling pathway increased and there were LCK、LAT、SYK、PKC expressed differently. Stripes of DNA amplification were founded in LCK、LAT、SYK、PKC while no stripe was showed in RASA1 in Idiopathic Orbital Inflammatory Pseudotumor. There was no stripe of DNA amplification in orbital cavernous hemangioma. LCK、LAT、SYK、PKC were positive expression in Idiopathic Orbital Inflammatory Pseudotumor. The expression of LCK、LAT、SYK、PKC was normal in orbital cavernous hemangioma. Positive stripes were showed in LCK、LAT、SYK、PKC Idiopathic Orbital Inflammatory Pseudotumor. There was no stripe in orbital cavernous hemangioma. Conclusion Over-expression of LCK、LAT、SYK、PKC in PI3K signaling pathwaymay take part in the pathogensis of Idiopathic Orbital Inflammatory Pseudotumor.%目的 分析特发性眼眶炎性假瘤患者与单纯眼眶海绵状血管瘤患者病变泪腺标本组织中LCK、LAT、SYK、PKC的差异表达情况,为特发性眼眶炎性假瘤发病机理提供参考.方法 采用全基因组表达谱芯片技术检测特发性眼眶炎性假瘤患者病变泪腺组织与单纯眼眶海绵状血管瘤患者瘤体组织中信号通路的差异性基因表达.采用PCR扩增技术,免疫组织化学染色方法及Westernblot法检测特发性眼眶炎性假瘤患者病变泪腺组织中 LCK、LAT、SYK、PKC的mRNA及蛋白质表达情况,并以单纯眼眶海绵状血管瘤患者瘤体组织标本作对照.结果 经基因芯片检测分析,实验组与对照组相比较,PI3K信号通路(PI3K signaling pathway)表达显著上调,其中LCK、LAT、SYK、PKC 4个基因差异性表达最为明显(P
