艳红桃芽变品系彩虹的RAPD分析

     

摘要

Three samples of the mutation shoots(YC0), first generation clone of the mutation shoot (YC1 ) and normal shoots (YY) in Yanhong peach were analyzed by RAPD to affirm the character hereditability of bud mutation further. The results showed that 110 bands were amplified from 13 random primers selected from 20 primers. 8. 5 bands were averagely amplified from one primer. The polymorphism band amplified from LC1 7 primer could be used as the RAPD marker to identify two varieties. The genetic distance between YC0(or YC, )and YY was 0. 0183. There was difference in DNA between YC0(or YC1) and YY but the difference was little.%为进一步确认芽变性状的可遗传性,对桃品种艳红自然变异原株的变异枝(YC0)及其无性系第一代(YC1)、艳红自然变异原株的正常枝(YY)2个品种(系)3个样品进行了RAPD分析.结果表明:从20个引物中筛选出13个随机引物,其中,共扩增出条带110条,平均每个引物扩增8.5条,其中引物LC17扩增出多态性条带,可用作鉴定这2个品种(系)的RAPD标记.YC0(或YC1)与YY的遗传距离(GD)为0.018 3.艳红与彩虹存在DNA水平的遗传差异,而特异多态性带较少与供试的2个品种性状差异较小一致.
