根结线虫天敌真菌1号分离物碱性磷酸酶的功能团分析

     

摘要

Effects of metal ions, organic reagents and chemical modifying agents on activity of alkaline phosphatase purified from DFRKN-1 were analyzed to know more about characters of alkaline phosphatase and provide reference for further study of the mechanism of NFRKN-1 invading into knot nematode and its development and utilization. The results showed that Mg2+ ,Ba2+ and K+ of certain concentrations activated the enzyme significantly, Zn2+ could inhibit the enzyme activity; Methanol, Ethanol and ethylene glycol inhibited the enzyme activity in similar degrees. Histidine residue and Lysine residue are essential groups of the alkaline phosphatase; sulfhydryl of cysteine was not an essential group, but disulfide bond played an important role in maintaining the activated conformation of alkaline phosphatase.%为进一步了解碱性磷酸酶的特性,为深入研究DFRKN-1侵入线虫的机理及开发利用提供借鉴,研究了金属离子、有机溶剂和化学修饰剂对根结线虫天敌真菌1号分离物(Destroying fungi of root-knot nematode-1,DFRKN-1)碱性磷酸酶酶活力的影响.结果表明:Mg2+、Ba2+、K+在一定浓度下对该酶有明显的激活作用,Zn2+对该酶有抑制作用;甲醇、乙醇、乙二醇对该酶均有明显的抑制作用,且抑制程度相当;酶的必需功能团有组氨酸残基、赖氨酸残基等;半胱氨酸的疏基不是酶活性的必需基团,但二硫键在维持酶活性构象中有重要作用.
