薏苡愈伤组织的诱导与分化

         

摘要

To provide scientific basis for the detoxication and rapid propagation of the fine variety of C. Lachrymal-jobi and the production of virue-free plantlets in plant area, the effects of different concentrations of mixed 2, 4-D, KT, NAA on callus induction, differentiation and seedling was researched. The materials of this research were aerial root of C. Lacryma-jobi, seed embryo and spears. The results showed that KT was the main factor affecting the differentiation of C. Lacryma-jobi spears and the generating of the virus-free plantlets. Aerial root was the best explants for inducing embryonic callus and embryoid and its optimal differentiation culture condition was MS + 2, 4-D 0. 4 mg/L + KT 1. 5 mg/L+NAA 0. 8 mg/L, its seedling culture medium was MS +2,4-D 0. 4 mg/L+ KT 1. 5. Mg/L+ NAA 1. 0 mg/L.%为薏苡优良品种的脱毒快繁和基地化脱毒种苗的生产提供科学依据,以薏苡气生根、种子胚、嫩茎为材料,研究了2,4-D、KT、NAA不同浓度配比对愈伤组织的诱导、分化和成苗的影响.结果表明,KT是薏苡芽分化及脱毒种苗生成的主要影响因素,气生根是诱导胚性愈伤组织及胚状体最合适的外植体,其最佳分化培养条件为MS+2,4-D0.4mg/L+ KT1.5mg/L+ NAA0.8mg/L,最佳成苗培养基为MS+2,4-D0.4mg/L+KT1.5 mg/L+NAA 1.0 mg/L.
