为筛选具有漆酶活性及对常用染料高效降解的菌株,对土壤样品进行富集培养筛选具有漆酶活性的菌株,利用形态学、生理生化特性和分子生物学方法确定菌株的分类地位,并研究菌株的生长特性及菌株芽孢漆酶对染料的脱色效果.结果表明:菌株HL2为芽孢杆菌属(Bacillus)细菌,其在25~55℃条件下能正常生长,在pH 5~10条件下生长旺盛;以干重计算,菌株HL2芽孢漆酶的酶活力为37.58 U/g;在pH为7.0的体系中,菌株HL2的芽孢漆酶对结晶紫和茜素红在6h内的脱色率分别为83.9%和81.60%;加入乙酰丁香酮(Ace)作介体时,对活性黑5的脱色率由48.95%提高至86.95%.%Strain Hl2 with a laccase activity is screened from soil samples after enrichment culture and the taxonomic status of Strain Hl2 is determined by morphology,physiological and biochemical characteristics and molecular biological methods.The growth characteristics and decoloring effect of spore laccase of Strain Hl2 on common dyes are studied to screen the bacteria strain with a laccase activity and highly effective degradation to common dyes.Results:Strain Hl2 belongs to Bacillus genus,grows normally at 25~55Cand grows vigorously within pH 5~10.The spore laccase enzyme activity per gram (dry weight) is 37.58 U.The decoloring rate of spore laccase of Strain Hl2 at pH 7.0 to crystal violet and alizarin red 83.9% and 81.60% within 6h respectively.When Acetosyringone is used to a vector,the decoloring rate of spore laccase of Strain Hl2 to Reactive black 5 can increases from 48.95% to 86.95 %.
