贵州地方辣椒材料疫病抗性鉴定的分子标记引物筛选

         

摘要

In order to provide references for identification on pepper phytophthora blight resistance and screening the molecular markers primer of local pepper in Guizhou,the resistance material S016(P1),high sensitive material S010(P2),S022(P3) and their hybrid F1 and F2 generation were used for the test material to report molecular marker primers related to phytophthora blight,and to screen molecular markers primers to disease resistance in pepper.Results:In the 18 candidate primers,16 pairs of primers were not specific in the three parents and their hybrid F1 DNA amplified bands.Moreover,Primer 3 was not specific in the three parent materials DNA amplified bands.So the 17 pairs of primers were not suitable for identification of resistance of local pepper resources blight in Guizhou.Primer 1 was specific in S016 and S022 parent materials DNA amplified bands.Primer 1 was used as the identification of disease resistance similar to S016 and S022 parents and their hybrids materials by verifying with hybrid F2 materials.%为贵州地方辣椒种质资源疫病抗性鉴定及抗疫病辣椒品种的分子标记辅助育种提供参考,以具有代表性的贵州地方辣椒抗疫病纯系母本S016 (P1)、高感疫病纯系父本S010(P2)和S022 (P3)及其杂交F1代和F2代为试验材料,用已报道辣椒疫病抗性鉴定相关分子标记的18对引物为候选标记引物,采用育苗接种鉴定和分子标记鉴定相结合的方法对适合贵州地方辣椒品种资源疫病抗性鉴定的分子标记引物进行筛选.结果表明:在18对候选引物中有16对引物对3个亲本及其F1代材料的DNA扩增条带无特异性,Primer3引物对3个亲本材料的DNA扩增条带无特异性,该17对引物均不适合用于贵州地方辣椒品种资源疫病的抗性鉴定.Primer1引物对S016和S022亲本材料的DNA扩增条带存在特异性,经F2代群体材料进一步验证,该引物可作为类似S016和S022亲本及其杂交后代材料的疫病抗性鉴定.
