利用紫外光谱研究麦冬蛋白质提取分离的优化条件

         

摘要

采用4℃下以不同的料液比、提取时间、磷酸盐缓冲溶液pH、乙醇溶液浓度、硫酸铵溶液浓度,进行单因素试验,对麦冬蛋白质浸提,测定蛋白质提取液的吸光度,确定了麦冬蛋白质提取分离的最优条件。研究结果表明:提取分离的优化条件是以料液比1:20,pH9.00的磷酸盐缓冲溶液提取6h。%Optimal conditions of extraction separation of Ophiopogon japonicus protein were studied by signal factor experiments of extracting protein under 4℃, including solid - liquid ratio, extracting time, pH of sodium phosphate buff- er, concentration of ethanol and ammonium sulfate, and the absorbance of extracting solution was determined. Results showed that the optimal separation conditions were extracting for 6 h at pH 9.00 in sodium phosphate buffer with the solid - liquid ratio of 1 : 20, respectively.
