基质金属蛋白酶-19在鼻咽癌组织的表达及临床意义

摘要

目的 探讨基质金属蛋白酶-19(MMP-19)在鼻咽癌组织中的表达及其与鼻咽癌发生发展的关系 . 方法 选择鼻咽癌组织99例(鼻咽癌组)、癌旁非肿瘤上皮组织24例(癌旁组)、正常鼻咽黏膜组织20例(正常组) ,运用组织芯片技术制成鼻咽癌组织芯片,采用免疫组化检测标本中MMP-19的表达. 结果 3组MMP-19的表达情况比较,差异有统计学意义(P<0.01),鼻咽癌组MMP-19失表达和低表达比例均明显高于另两组(P<0.01);鼻咽癌组织MMP-19表达与性别、组织学分类、淋巴结转移、临床TNM分期无明显关系( P>0.05). 结论 MMP-19在大部分鼻咽癌细胞表达下调,示其可能参与了鼻咽癌发生过程,但MMP-19在鼻咽癌中的具体作用和机制尚未清楚,仍待进一步深入研究.%Objective To investigate the expression of matrix metalloproteinase(MMP)-19 in nasopharyngeal carcinoma(NPC) tissues and its relationship with the occurrence and development of NPC.Methods The samples were collected from 99 cases of nasopharyngeal carcinoma(NPC group),24 cases of adjacent nontumorous epithelial tissue (parac-arcinoma tissue group) and 20 cases of normal nasopharyngeal mucosa( normal group) to gain the tissue microarray of NPC by tissue microarray technique .The expression of MMP1-9 in the samples was detected by immunohistochemistry.Result s There was significant difference in the MMP-19 expression among three groups(P<0.01). The proportion of loss of MMP-19 in NPC group was significantly higher than that in the other two groups,as well as the proportion of low expression of MMP-19(P<0.01).The expression of MMP-19 in NPC tissues did not significantly correlate with gender,histological classification, lymph node metastasis,clinical TNM stage(P>0.05).Conclusion The down-regulated expression of MMP-19 can be observed in most NPC cells,which suggests that it might involve in the process of NPC development.Howevert,he role and mechanism of MMP-19 in of NPC are not confirmed,and further study is needed.
