阿尔茨海默病患者Ghrelin水平及其与Toll样受体4信号通路的关系

             

摘要

目的 探讨阿尔茨海默病(AD)患者的Ghrelin水平及其与Toll样受体4(TLR4)信号通路的关系.方法 选择AD患者和健康人群各60例分别作为AD组及对照组.比较两组外周血中Ghrelin水平、外周血单核细胞膜上TLR4阳性表达率及平均荧光强度(MFI)、蒙特利尔认知评估量表(MoCA)评分.分析AD病患者Ghrelin水平与TLR4阳性率及MFI、MoCA评分的相关性.结果 AD组的外周血Ghrelin水平及MoCA评分均低于对照组(均P<0.05),而TLR4阳性率、MFI均高于对照组(均P<0.05).AD组患者外周血Ghrelin水平与TLR4阳性率及MFI均呈负相关(均P<0.05),而与MoCA评分呈正相关(P<0.05).结论 AD患者的外周血Ghrelin水平下降,且认知功能损害越重Ghrelin水平越低,其或可通过抑制TLR4介导的信号通路减轻AD患者的免疫炎症反应.%Objective To explore the Ghrelin level and its relation with Toll-like receptor-4 ( TLR4 ) signal pathway in patients with Alzheimer's disease( AD) . Methods Sixty patients with AD and 60 healthy subjects were enrolled as AD group and control group respectively. The Ghrelin level of peripheral blood,positive expression rate and median fluorescence intensity( MFI) of TLR4 of peripheral blood mononuclear cells,and Montreal Cognitive Assessment( MoCA) score were compared between the two groups. The correlation of the Ghrelin level with the positive expression rate and MFI of TLR4 and MoCA score was analyzed in the AD patients. Results Compared to the control group,the Ghrelin level of peripheral blood and MoCA score were lower but the positive expression rate and MFI of TLR4 were higher in the AD group(all P<0. 05). In the AD group,the Ghrelin level of peripheral blood negatively correlated with the positive expression rate and MFI of TLR4(all P<0. 05),but positively correlated with MoCA score(P<0. 05). Conclusion The Ghrelin level of peripheral blood decreases in the AD patients,which is lower when cognitive impairment is more severe. And it might reduce the immune inflammatory response in the AD patients by inhibiting the TLR4-mediated signaling pathway.
