Objective To estahlish a rat model of pulmonary hypertension secondary to left - to - right shunt and to study the correlation between BMPR Ⅱ and apoptosis of pulmonary artery smooth muscle cells ( PASMCs ). Methods Forty SD rats were randomly divided into 4 groups: shunt 6 weeks group, non - shunt 6 weeks group, shunt 11 weeks group and non - shunt 11 weeks group. Left - to - right shunt rat model was constructed by shunting abdominal aorta to inferior vena cava. TUNEL and Western blot were applied for detection of nuclear DNA fragment from apoptotic cells and BMPR Ⅱ protein expression, respectively. Results With the prolongation of shunt, significant down - regulation of BMPR Ⅱ protein and reduction of PASMC apoptosis were revealed in these rats. Conclusion In rats with left - to - right shunt - induced pulmonary hypertension, simultaneous down - regulation of BMPR Ⅱ and reduction of apoptosis of PASMCs suggests their close association.%目的 建立左向右分流所致肺动脉高压的大鼠模型,探讨模型大鼠肺组织中骨形态生成蛋白受体Ⅱ(BMPRⅡ) 变化与肺动脉平滑肌细胞凋亡的关系.方法 40 只 SD 大鼠随机分为 4 组:分流 6 周组、对照 6 周组、分流 11 周组及对照 11 周组.采用腹主动脉-下腔静脉分流术建立左向右分流的肺动脉高压大鼠模型,用原位缺口末端标记(TUNEL)方法对大鼠肺血管平滑肌细胞凋亡状态进行观察,用 Western blot 法检测肺组织平滑肌细胞中 BMPRⅡ的表达变化.结果 随着分流时间的延长,大鼠肺组织中 BMPRⅡ 的表达减少,而此时肺组织血管平滑肌细胞凋亡也明显减少.结论 左向右分流肺动脉高压大鼠肺血管平滑肌细胞凋亡减少,可能与 BMPRⅡ表达下调有一定关系.
