荔枝核总黄酮抗大鼠肝纤维化的作用及其对核转录因子-κB p65表达的影响

             

摘要

目的 观察核转录因子-κB(NF-κB)p65在实验性大鼠肝纤维化肝组织中的表达,探讨荔枝核总黄酮(TFL)抗肝纤维化的作用机制.方法 以二甲基亚硝胺腹腔注射制作大鼠肝纤维化模型;造模同时,模型组以生理盐水、TFL组以TFL[200 mg/(kg·d)]、秋水仙碱组以秋水仙碱灌胃给药;6周后处死大鼠,抽取下腔静脉血检测AST、ALT,取肝脏同一部位行HE染色观察大鼠肝纤维化程度,采用免疫组化检测各组肝组织NF-κB p65的表达.结果 TFL组和秋水仙碱组血清AST、ALT均明显低于模型组,差异有统计学意义(P<0.05).与模型组比较,TFL组NF-κB p65的表达明显抑制,大鼠肝纤维化程度改善(P<0.05).结论 TFL可减轻肝损伤及其纤维化程度,抑制NF-κB p65的表达,这可能是其抗肝纤维化作用的机制之一.%Objective To investigate the effects of total flavone from Litchi chinensis Sonn (TFL) on the expression of NF - κB and hepatic fibrosis in rats. Methods The rat model of hepatic fibrosis was established by intraperitoneal injection of dimethylnitrosamine. TFL, colchicine and saline lavage were given to the subjects in the TFL, model and col-chicine groups, respectively. Serum ALT and AST were measured at 6th week, and the pathological observation was also performed on left liver lobes with HE straining while S - P immunohistochemical staining using polyclonal antibodies a-gainst NF - κB was applied in assessment of NF - κB. Results The serum ALT and AST in TFL and colchicine groups were significantly lower than that in model group ( P < 0. 05 ) . Meanwhile, significant improvement of hepatic fibrosis and suppression of in NF - κB were revealed in TFL group than that in model group ( P < 0. 05 ) . Conclusion TEL alleviates the hepatic fibrosis in rats, which may involve the inhibition of NF - κB.
