超声辅助贯叶连翘中总黄酮的纯化工艺研究

         

摘要

贯叶连翘提取物及其制剂是北美和欧洲最畅销的抗抑郁植物药,大量临床研究证实其对中,轻度抑郁症有效,且副作用较传统的化学抗抑郁药轻微。新的研究表明,其有效成分具有抗病毒,抗癌抗肿瘤以及治疗肝炎,白血病等多种药理作用。文章综合运用超声波提取法,通过单因素实验,分别考察了提取溶剂的用量,提取溶剂的浓度,提取温度,提取时间对提取效率的影响。再以提取溶剂的用量,提取溶剂的浓度,提取温度,提取时间四因素为指标,设计正交实验。研究结果表明影响贯叶连翘中黄酮提取的主次因素为提取溶剂的浓度〉提取温度〉提取时间〉提取溶剂的用量,其最佳提取工艺条件为用10倍用量的50%乙醇,在50℃温度下提取40 min,在该条件下提取贯叶连翘所得黄酮含量为15.8%。优选出的最佳提取工艺条件,为开发贯叶连翘中抗抑郁有效成分提供了一个科学、合理的理论依据。%ypericum perforatum extracts and their preparation are the most popular antidepressant plant drugs in north America and Europe.It is effective for the treatment of mild to moderate depression,which has been verified by numerous preclinical and clinical studies.Fewer side effects have been detected compared with traditional antidepressants.It is showed by newly study that its active principle more and more pharmacology effects has been discovered lately,such as antivirus,anti-cancer,hepatitis healing,leucocythemia healing. The paper comprehensively uses a variety of extraction and separation and purification technology,aiming to develop and take best advantage of hypericum perforatum.Ultrasonic extraction method,taken flavonoids yield as index,the effects of extraction solvent consumption,extraction solvent concentration,extraction temperature and extraction time on total flavonoids yield were investigated separately by single factor test.Orthogonal test was designed by extraction solvent consumption,extraction solvent concentration,extraction temperature and extraction time.The result show that factors affecting flavonoid yield was in the order of ethanol concentration,extraction temperature,extraction time and ethanol consumption.Through this experiment we found out the best condition of extracting the flavonoids from Hypericum perforatum is the ethanol concentration of 50 %,ethanol consumption 10 times,extraction temperature 50 ℃,extraction time 40min measured flavonoids content reached 15.8 %.Process condition is optimized,which forms the scientific and reasonable bases for developing the anti-impressive active parts from Hypericum pelforatum.
