黄芪-丹参对脂多糖致急性肺损伤大鼠的防治作用

             

摘要

Objective To investigate the therapeutic effect and mechanism of Astragalus and Salvia miltiorrhiza Extract on acute lung injury rats induced by lipopolysaccharide. Methods 72 male SD rats were randomly divided into control group (the same amount of saline intervention), the model group (the same amount of saline intervention), the positive control group (dexamethasone 5 mg / kg) and the experimental group (0.59 g / mL, Astragalus - Salvia extract, 2 mL) , 18 rats in each group, the model group, positive control group and experimental group were modeled by LPS. Results The pathological scores and lung wet / dry ratio in the model group were significantly higher than those in the control group ( P<0.05);The pathological scores of lungs and the lung tissue wet / dry ratio in the positive control group and the experimental group were significantly lower than those in the model group (P<0.05); 72h after modeling, PaO2 , pH in the model group were significantly lower than those in the control group ( P<0.05);PaO2 and pH in the positive control group and experimental group were significantly higher than those in the model group ( P<0.05);72h after modeling, TNF-αin the lung in model group was sig-nificantly higher than that in the control group, TNF-α, IL-1 beta and IL-6 in positive control group and experimental group were significantly higher than those in the control group;72h after modeling, ICAM-1, kappa B NF-( p65) mRNA in the lung tissue of positive control group and the experimental group were sig-nificantly lower than those of model group (P<0.05).Conclusion The effects of Astragalus and Salvia miltiorrhiza extract on ALI rats induced by lipopolysaccharide ( LPS) were significant, which could signifi-cantly reduce the level of inflammatory response and down regulate the expression levels of ICAM-1 and NF-( p65) in lung tissue.%目的 探讨中药材黄芪-丹参提取物对脂多糖所致急性肺损伤大鼠治疗作用及其机制.方法 选取72只雄性SD大鼠,随机分为对照组(等量生理盐水干预)、模型组(等量生理盐水干预)、阳性对照组(地塞米松5 mg/kg)和实验组(0.59 g/mL浓度的黄芪-丹参提取物2 mL灌胃)各18只,模型组、阳性对照组和实验组采用LPS造模.结果 模型组大鼠的肺组织病理学积分、肺组织湿/干比均显著的高于对照组(P<0.05);阳性对照组和实验组大鼠的肺组织病理学积分、肺组织湿/干比均显著的低于模型组(P<0.05);造模72小时后,模型组大鼠的PaO2、pH值均显著低于对照组(P<0.05);阳性对照组和实验组PaO2、pH值均显著高于模型组(P<0.05);造模72小时后,模型组大鼠的肺组织中TNF-α均显著高于对照组(P<0.05);阳性对照组和实验组肺组织中TNF-α、IL-1β、IL-6均显著的低于模型组(P<0.05);造模72小时后,阳性对照组和实验组肺组织ICAM-1、NF-κB(p65)mRNA水平均显著的低于模型组(P<0.05).结论 黄芪-丹参提取物对脂多糖所致ALI大鼠治疗效果显著,能显著减轻炎性反应程度、下调肺组织中ICAM-1、NF-κB(p65)的表达水平.
