HPLC法测定四酸粉中水杨酸和苯甲酸的含量

         

摘要

To establish an HPLC method to determine salicylic acid and benzoic acid in four acid powder . Methods: A C18 column (150 × 4 . 6mm , 5μm) was used , the column temperature was set at 25℃, the mobile phase was consisted by 0 . 05mol/L potassium dihydrogen phosphate (adjusted to pH 3 . 2 with phosphoric acid)- methanol (60:40), and flow rate was 1 . 0 ml/min , the detection was conducted at wavelength of 225 nm . Results:The calibration curves were linear in the range of 14 . 226~33 . 194μg/ml(r=0 . 9998)for salicylic acid and 7 . 278~16 . 982μg/ml(r=0 . 9999) for benzoic acid . The average recovery of salicylic acid was 100 . 6% with RSD of 0 . 50%, and the average recovery of benzoic acid was 100 . 6% with RSD of 0 . 15%. Conclusion:This proposed method appeared to be simple , rapid and accurate , and can be used to control the product quality and preparation process .%目的:建立HPLC法测定四酸粉中水杨酸和苯甲酸的含量。方法:采用C18色谱柱(150×4.6mm,5μm),以0.05mol/L磷酸二氢钾溶液(用磷酸调节pH至3.2)-甲醇(60:40)为流动相,柱温25℃,流速为1.0mL/min,检测波长为225nm,进样量为20μL。结果:线性范围分别为水杨酸14.226~33.194μg/mL,r=0.9998,苯甲酸7.278~16.982μg/mL,r=0.9999;平均回收率为水杨酸100.6%(RSD=0.50%),苯甲酸100.6%(RSD=0.15%)。结论:本文方法简便,快速,准确,可作为本品质量评价和生产工艺监控的有效方法。
