垂花蕙兰丛生芽途径的组织培养技术研究

     

摘要

Lateral buds were used as the explants for the study on tissue culture of pendent Cymbidium .It showed that by using 72% ethanol ,0.1% mercuric chloride and 15% sodium hypochlorite to disinfect the lateral buds three times ,the bud survival rate could be as high as 56% .The disinfection was especially effective in springtime .The induction rate of the clustered shoots was up to 100% with numerous differentiated buds when 6-BA was applied at the rate of 4.0 mg · L -1 .During the proliferating period of the clustered shoots ,the 6-BA treatment was a key factor that produced the best effect with a proliferating coefficient of clustered shoots of 3.4 and strong buds .For the development in the rooting stage ,on the other hand ,a 1/2 MS medium containing NAA 2.0 mg · L -1 + banana slurry 50 g · L -1 +AC 1.0 g · L -1 appeared to be most desirable .And ,moss seemed to be the best base material for the cultivation of the pendent Cymbidium plantlets .%以侧芽为外植体,对垂花蕙兰丛生芽途径的组织培养进行研究,结果表明,采用72%乙醇、0.1%升汞和15%次氯酸钠对侧芽进行3次消毒,存活率高达56%,尤其是在春季外植体消毒效果佳;在丛生芽诱导的过程中,6-BA浓度为4.0 mg · L -1时,丛芽诱导率可达100%且丛芽分化较多;在丛生芽增殖过程中,6-BA是影响垂花蕙兰丛生芽增殖的重要因素,6-BA 4.0 mg · L -1处理增殖系数为3.4,丛生芽较健壮,适合丛生芽增殖;培养基1/2MS+NAA2.0 mg · L -1+香蕉泥50 g · L -1+AC1 g · L -1生根壮苗效果较好;苔藓是垂花蕙兰试管苗移栽较好的基质。
