Purpose To investigate mechanisms of edema formation following intracerebral hemorrhage and the effects of hirudin. Methods The animals were injected fresh autologous caudal artery blood into the caudate nucleus without heparin by an oriented cannula. Changes in brain water and Na+、 K+、Ca2+、Mg2+ contents were measured at 6、12、24、48、72、144 h after ICH. The response by adding a specific thrombin inhibitor, hirudin to the brain was assessed. Results There are some relations between brain water and ion contents within the earlier time following intracerebral hemorrhage and hirudin reduces brain water and ion contents changes(P<0.05). Conclusions Because that hirudin reduces brain edema, thrombin may play an important role in mechanisms of edema formation following intracerebral hemorrhage.%目的 探讨自发性脑出血脑水肿的发生机制及水蛭素的作用。方法 利用大鼠自体新鲜无肝素未凝动脉血定量脑出血模型,观察不同时间大鼠脑出血周围脑组织含水量、离子含量的变化,并以大鼠脑出血后脑内注射凝血酶特异抑制剂水蛭素的实验,研究水蛭素对水肿程度及离子含量变化的影响。结果 大鼠脑出血后早期脑组织含水量、离子随时间推移而升高、变化,而水蛭素可减轻其水肿程度及离子变化(P<0.05)。结论 凝血酶与实验性脑出血脑水肿的产生发展关系密切,早期应用水蛭素可减轻脑出血脑水肿。
