响应面优化铁皮石斛须根多糖提取工艺研究

         

摘要

Dendrobium roots as raw material, the polysaccharide was extracted by ultrasonic assisted technique. On the basis of single factor experiment, the design principle of Box-Behnken center combination in response surface methodology (RSM) was applied to analyze and optimize the technological conditions such as liquid to liquid ratio, extraction time and extraction temperature. The results showed that the size of three factors as ex-traction temperature>solid-liquid ratio>extraction time, the optimal extraction conditions of solid-liquid ratio 1:29 (g/mL), extraction time 42 min, extraction temperature of 62℃, the rate of the actual value of 8.03%polysaccharides under this condition, the smaller the gap with the theoretical value of 8.14%. Showed that the regression model was reasonable and effective, to optimize the extraction process of polysaccharide of Dendrobi-um roots had very good guidance.%以铁皮石斛须根为原料,采用超声辅助技术提取多糖.在单因素试验的基础之上,利用响应面分析法中的Box-Behnken中心组合设计原理对料液比、提取时间、提取温度等工艺条件进行分析优化.结果表明:3个因素的影响大小为提取温度>料液比>提取时间,最佳提取工艺条件为料液比1:29(g/mL),提取时间42 min,提取温度62℃,在此条件下多糖得率实际值为8.03%,与理论值8.14%差距较小.表明该回归模型合理有效,对石斛须根多糖提取工艺的优化有很好的指导作用.
