GC法测定藿胆分散片中百秋李醇的含量

         

摘要

Objective To establish a method for determination of patchouli alcohol in Huodan Dispersible Tablets by gas chromatography (GC).Methods GC detection was performed after ultrasonic extraction and centrifugation. Capillary column with PEG-20M (30 m×0.25 mm, 0.5μm) as stationary phase was adopted. Column temperature was programmed: the initial temperature was 120℃ and was kept for 5 min; the temperature was elevated to 160℃ at the speed of 10℃/min and was kept for 11 min. The gasification temperature was 260℃ and the detector temperature was 280℃. The carrier gas was N2, with the flow rate of 3.69 mL/min; the split ratio was 30:1; the injection volume was 1μL, usingn-octadecane as an internal standard.ResultsThe calibration curve of patchouli alcohol was linear in the range of 5.64×10-3-2.256×10-2 mg/mL (Y=175.57X-0.036) with a correlation coefficient of 0.9990. The average recovery was 101.61 % (n=6).Conclusion The method is simple and of high sensitivity and reproducibility, which can be used as main method of quality control of Huodan Dispersible.%目的:建立毛细管气相色谱法(GC)测定藿胆分散片中百秋李醇含量的方法。方法样品经超声提取、离心过滤后采用GC测定。色谱条件:色谱柱为PEG-20M为固定相的毛细管柱(30 m×0.25 mm,0.5μm);柱温采用程序升温,初始温度120℃,保持5 min,再以10℃/min的速率升温至160℃,保持11 min;进样口温度260℃;FID检测器,温度280℃;载气为氮气,流速3.69 mL/min,分流比30:1;进样量1μL。以正十八烷为内标物。结果百秋李醇在5.64×10-3~2.256×10-2 mg/mL浓度范围内呈良好的线性关系(Y=175.57X-0.036,r=0.9990),平均回收率为101.61%(n=6)。结论该方法简便、灵敏度较高、重现性良好,可作为藿胆分散片质量控制的主要方法。
