HPLC-DAD-MS/MS法分析金银花中化学成分

         

摘要

Objective To analyze the chemical constituents in Lonicerae japonicae Flos by HPLC-DAD-MS/MS. Methods The HPLC analysis was performed on a Hypersil C18 column (250 mm×4.6 mm, 5 μm). A mixture of solvent A (0.1%aqueous solution of formic acid) and solvent B (acetonitrile containing 0.1%formic acid) was used as the mobile phase at a flow rate of 1.0 mL/min, and the scanning wavelength was 200-400 nm for DAD. Multi-stage ESI-MS was adopted to analyze the chemical constituents in Lonicerae japonicae Flos, under both positive and negative ion detection modes. The structure was identified based on MS/MS fragment ion assignment, together with the data derived from reference substance and literature. Results Seventeen major constituents including six organic acids, three flavonoids and eight iridoids were identified. Conclusion The method is simple, rapid and sensitive, which can be used in the analysis of chemical constituents in Lonicerae japonicae Flos.%目的:采用高效液相色谱-二极管阵列检测器(HPLC-DAD)和液相色谱-质谱联用(LC-MS/MS)法分析金银花中的化学成分。方法 Hypersil C18色谱柱(250 mm×4.6 mm,5μm),流动相A为水(0.1%甲酸),B为乙腈(0.1%甲酸),梯度洗脱,流速1.0 mL/min,DAD扫描波长范围为200~400 nm。质谱采用电喷雾离子源(ESI),正、负两种离子检测模式同时对金银花中化学成分进行多级质谱裂解分析。以对照品及文献数据为对照,通过解析各成分的MS/MS谱图,指认各成分。结果共鉴定了17个金银花中的主要成分,包括6个有机酸类成分,3个黄酮类成分,及8个环烯醚萜。结论此法操作简单、快速、灵敏,适用于金银花中化学成分的分析和结构鉴定。
