目的:分析研究人胎盘来源间干细胞(hPDSCs)在妊娠期糖尿病(GDM)脂代谢中的作用。方法:采用贴壁分离筛选法获取hPDSCs。30只SD孕鼠随机分成3组,每组10只。实验组为植入hPDSCs的GDM孕鼠;对照组为GDM孕鼠;空白组为正常孕鼠,不注入任何试剂。采用酶联免疫吸附试验(ELISA)法检测抵抗素及脂联素在各组大鼠血清中的表达。结果:实验组抵抗素水平低于对照组,分别为(4.43±0.04)和(10.57±0.12)mg/L,差异有统计学意义(t=153.500,P<0.01);实验组脂联素水平高于对照组,分别为(11.98±1.20)和(5.58±0.67)mg/L,差异有统计学意义(t=14.726,P<0.01)。结论:hPDSCs对GDM的进展有一定干预作用。%Objective: This study aimed to analyze the effect of human placenta-derived stem cells on lipid metabolism of gestational diabetes mellitus.Methods:Human placenta-derived stem cells were harvested with adherent separation and screening method. Thirty Sprague-Dawley pregnant rats were randomly divided into three groups, with ten rats in each group. Experimental group: human placenta-derived stem cells were implanted into pregnant rats with gestational diabetes mellitus. Control group: pregnant rats with gestational diabetes mellitus. Blank group: normal pregnant rats, without any injection. The expression of resistin and adiponectin in the serum of rats in each group was detected with enzyme-linked immunosorbent assay method. Results: The resistin expression levels in experimental group were lower than that in control group (4.43±0.04 mg/L, 10.57 ± 0.12 mg/L), the difference was statistically signiifcant between the two groups (t=153.500,P<0.01). The adiponectin expression levels in the experimental group was higher than that in control group, (11.98±1.20 mg/L, 5.58±0.67 mg/L), the difference was statistically signiifcant between the two groups (t=14.726,P<0.01).Conclusion:Human placenta-derived stem cells have certain intervention effects on the progression of gestational diabetes mellitus.
