大鼠肝癌免疫微环境中Treg与IL-10水平的变化及意义

         

摘要

目的:探讨大鼠肝癌免疫微环境中Treg的数量与IL-10表达水平的变化及意义。方法40只雄性Wistar大鼠随机分成三组,肝癌模型组(20只)、生理盐水组(10只)、空白对照组(10只),肝癌模型组大鼠按100 mg/kg给予腹腔注射DEN(二乙基亚硝胺)诱导肝癌模型。光镜下观察肝组织病理改变,流式细胞术检测肝组织CD 4+CD 25+Treg的百分率,应用免疫组织化学SABC法检测肝组织Treg和IL-10的表达水平,并对Treg和IL-10的变化及意义进行研究。结果成功制备大鼠诱导性肝癌模型,在镜下观察到纤维化、癌结节和癌细胞形成。流式细胞术检测显示肝癌模型组肝癌组织中Treg占CD 4+T细胞的比例为(14.32±4.84)%,显著高于空白对照组的(5.64±6.10)%和生理盐水组的(7.95±3.55)%,差异有统计学意义(P<0.01)。免疫组化法检测肝癌模型组肝癌组织中Treg和IL-10的表达高于空白对照组和生理盐水组(P<0.05)。CD 4+CD 25+Treg与IL-10的水平呈明显正相关(P<0.05)。结论大鼠肝癌免疫微环境中IL-10和Treg相互作用,共同参与了对肝癌免疫微环境的免疫抑制调节作用。%Objective To discuss the Changes and significance of the number of Treg cell and the expression level of IL-10 in the immune microenvironment of rat liver cancer. Methods 40 male Wistar rats were randomly divided into three groups, liver cancer model group (n=20), saline group (n=10) and blank control group (n=10). Liver cancer model rats receive intraperitoneal injection of DEN(100 mg/kg) to induce dliver cancer model.Observe the pathological changes of the liver tissue by light microscope.Detecte the percentage of CD 4+CD 25+Treg cell in iver tissue by fiow cytometry.Detecte the expression level of Treg cell and IL-10 in liver tissue by immunohistochemical SABC. Then analysis the Changes and significance of Treg cells and IL-10. Results We successfully induced rat liver cancer model.We observed the cancer cells and cancer nodules under the microscope. The consequence of flow cytometry detection showed the ratio of Treg cells in CD 4+T cells was (14.32 ± 4.84)%in liver cancer model group, significantly higher than the blank control group (5.64 ± 6.10)%and the saline group (7.95 ±3.55)%.The difference was statistically significant (P<0.01). The consequence of immunohistochemical detection showed the ratio of the express lever of Treg cells and IL-10 in liver cancer model group was significantly higher than the blank control group and the saline group (P<0.05). The variance of CD 4+CD 25+Treg cells was significantly positively correlated with IL-10(P<0.05). Conclusion IL-10 and Treg cells in rat hepatoma immune microenvironment interact in the immune microenvironment of liver cancer immunosuppression.
