大鼠跟腱损伤修复局部生长因子内源性表达变化的研究

         

摘要

目的 测定损伤肌腱修复术后康复过程中手术局部分泌型生长因子表达的变化情况.方法 21只大鼠切断单侧跟腱后立即以丝线修复,术后第1、3、5、7、14、21和28天分别处死3只实验大鼠.收集损伤局部及健侧跟腱标本,以实时荧光定量PCR(qRT-PCR)测定转化生长因子β1(TGF-β1)、TGF-β2、TGF-β3,骨形态形成蛋白(BMP)-12、BMP-13、BMP-14,血管内皮生长因子(VEGF),肝细胞生长因子(HGF),组织连接生长因子(CTGF),胰岛素样生长因子1(IGF-1)、IGF-2,成纤维细胞生长因子(FGF-1)、FGF-2,血小板源性生长因子a(PDGFa)、PDGFb和表皮生长因子(EGF)等分泌型生长因子的表达水平并进行对比分析.结果 术后第1、3、5、7、14、21和28天损伤修复测标本,TGF-β2表达水平最高,其中术后第1、3、5天表达最明显.BMP-13、BMP-14,VEGF和IGF表达水平基本相当,但较TGF-β2表达水平少.术后第7天,上述因子表达水平显著减少.而TGF-β1在术后第21天达到最大表达.对照组标本未测出相关生长因子RNA表达.结论 TGF-β2、BMP-13、BMP-14、IGF-1和VEGF在术后第1天即开始高表达,术后第7天显著降低.TGF-β1在术后第21天表达水平达最大值.在肌腱损伤后局部综合应用上述生长因子可有效加快其愈合进程,增加肌腱修复后的强度,促进肌腱早期修复.%Objective To determine the change of expression of local secretive type of growth factors from injured tendons during rehabilitation course. Methods The Achilles tendon in 21 rats were cut off and immediately repaired with silk thread. 3 ratt were sacrificed on 1,3,5,7,14,21,28 d, respectively. The injured tendons were collected for quantitative real-time(qRT) PCR analysis of the expression pattern of growth factors including TGF-β1 ,TGF-β2 ,TGF-β3 ,BMP-12 ,BMP-13,BMP-14 ,VEGF, HGF,CT-GF,IGF-1 ,IGF-2,FGF-1,FGF-2,PDGFa,PDGFb,and EGF. Tissue from uninjured tendons was harvested as control. Results The values of TGF- 2 were highest on 1,3,5,7,14,21,28 d,especially on 1, 3 , 5 d. The expression levels of BMP-13 ,BMP-14 , VEGF and IGF were basically similar, but lower than that of TGF-β2. On 7 d after operation, the expressions of these factors were markedly reduced. TGF-1 had maximum expression on 21 d. No RNA expression of related GF could be extracted from the samples in the control group. Conclusion TGF-β2 , BMP-13 ,BMP-14 , IGF-1 ,and VEGF are highly expressed on 1 d after injury and significantly reduced on 7 d. TGF-β1 peaks on 21 d. Integrated local application of these GF after tendon injury may accelerate the healing process,increase tendon repair strength and promote early repair of tendon.
