基于聚硫堇/纳米金固定酶的过氧化氢生物传感器

     

摘要

将电子媒介体硫堇(Thi)聚合于玻碳电极(GC)表面形成带正电的多孔聚硫堇(PTH)复合膜,再利用共价结合和静电吸附将纳米金(nano-Au)和过氧化物酶(HRP)修饰于电极上,从而制得HRP/nano-Au/PTH/GC传感器.用循环伏安法和计时电流法考察该修饰电极的电化学特性,发现该修饰电极对过氧化氢(H2O2)的还原有良好的电催化作用.实验结果表明:该传感器对H2O2的线性响应范围为1.4×10-6~4.26×10-3mol L,线性相关系数R=0.9993(n=23),检测下线为4.0×10-7mol L(S N=3),并具有选择性好、灵敏度高、响应快等优点.%A new approach to measure hydrogen peroxide(H2O2) with HRP/nano-Au/ PTH /GC modified electrode has been developed.First,a stable poly-thione film was polymerization on glassy carbon electrode.Then nano-gold and horseradish peroxidase was successful immobilized onto the above prepared film to construct the hydrogen peroxide biosensor via the effect of electrostatic adsorption and process of covalent bonding.The performance of the biosensor was characterized by cyclicvoltammetry and the chronoamperometry.The results confirmed that the electrode exhibited high sensitivity,fast response,and linear response in the range of 1.4×10-6 molL~4.26×10-3 molL with correlation coefficients of 0.9993(n=23),respectively,and the detection limit of 4.0×10-7 molL(SN=3).
