凝胶滞留电泳法检测人宫颈鳞癌细胞核中ETS1结合活性

         

摘要

目的 应用人宫颈鳞癌组织细胞核蛋白提取物与特异ETS1基因探针序列结合,经凝胶滞留电泳(EMSA)分析其结合能力.初步探讨ETS1作为转录因子在人宫颈鳞癌发生、进展中的作用机制.方法 通过EMSA,应用探针标记技术,在转录水平进一步揭示了ETS1结合DNA 能力的调控作用.结果 随着宫颈鳞癌的发生发展,ETS1结合DNA的能力增强,EMSA中探针迁移率明显降低(P<0.01).结论 ETS1与宫颈癌发生发展有相关性,其与肿瘤浸润和转移有密切的关系.%Objective To investigate the role of ETS1 as a transcription factor in development and progression rnof human cervical cancer by binding ETS1 gene-specific probe to protein concentrate from cervical cancer on elecrntrophoretic mohility shift assay (EMSA). Methods Binding of ETS1 proteins to DNA was examined with specific rnprobes and EMSA, which would shed light on the regulatory activity of ETS1 at the level of transcription. Results rnEMSA showed higher DNA-binding ahility of ETS1 along with progression of cervical lesions. Conclusion ETS1 is rncorrelated with development and progression of cervical cancer, and is also closely associated with invasiveness of the rntumor.
