液相色谱-质谱联用法测定犬血浆中青蒿素浓度

         

摘要

Objective To establish a liquid chromatography-mass spectrometry (LC-MS) method for the deterrnmination of artemisinin in dog plasma. Methods Artemether (AMT) was used as the internal standard. Solution of rnacetonitrile and ammonium acetate at 10 mmol/L (containing 0.1% formic acid) (85:15, V/V) was used for mobile phase rnpreparation with a flow rate at 0.3 ml/min. An agilent-C18 column (50 mm×4.6 mm, 1.8 μm) was used for separation.rnElectrospray ionization (ESI) and multiple reaction monitoring (MRM) were used for positive ion detection. Results rnThe linear calibration curves were ohtained in the concentration range of 2~500 ng/ml, with the lowest limit of quantifirncation at 2 ng/ml. Both inter and intra day precision (RSD) were less than 7.7%. The extraction recovery was between rn78% and 90%. Conclusion The method is qualified for artemisinin pharmacokinetics study in dogs.%目的 建立犬血浆中青蒿素(ART)浓度的液相色谱-质谱联用方法.方法 以蒿甲醚(AMT)为内标,采用乙腈和10 mmoL/L醋酸铵(含0.1%甲酸)溶液(85∶15,V/V)为流动相,流速为0.3 ml/min,Agilent-C18(50 mm×4.6 mm,1.8 μm)色谱柱为分析柱,采用电喷雾电离源(ESI),多反应监测方式进行正离子检测.结果 测定犬血浆中青蒿素浓度的线性范围为2~500 ng/ml,定量下限为2 ng/ml.日内、日间精密度(RSD)均<7.7%,提取回收率78%~90%.结论 本方法适应于犬体内青蒿素药代动力学研究.
