RhoC在人脑胶质瘤中的表达及其临床意义

         

摘要

目的 研究RhoC在人脑胶质瘤组织中的表达情况及其与病理分级的相关性,初步探讨RhoC在胶质瘤的诊断、恶性程度及预后判断中的临床意义.方法 采用抗亲和素-生物素复合物法(SABC)免疫组织化学法、反转录聚合酶链反应(RT-PCR)法对62例不同病理级别的星形细胞瘤及11份正常脑组织标本进行RhoC蛋白和RhoC mRNA的检测.结果 ①免疫组织化学结果显示62例胶质瘤组织中RhoC蛋白表达全部呈阳性,对照组正常脑组织存在弱表达或不表达.②不同病理级别星形细胞瘤标本的RhoC阳性细胞率、RhoC mRNA与甘油醛-3-磷酸脱氢酶(GAPDH)mRNA灰度比值均存在差异;Ⅰ级、Ⅱ级和Ⅲ、Ⅳ级之间差异有统计学意义(P<0.05),Ⅲ、Ⅳ级RhoC mRNA表达均高于Ⅰ级、Ⅱ级.③相关性分析:RhoC阳性细胞率和RhoC mRNA表达量与胶质瘤病理分级均呈正相关性(r=0.814,r=0.863,P 均<0.01).结论 RhoC可以作为星形细胞瘤Ⅰ~Ⅳ级新的有效的诊断标记物,同时作为常规组织病理学的分级标准的补充.%Objective To investigate RhoC expression in human glioma tissues and the correlation with pathological grading, and explore the implications for diagnosis, malignancy and prognosis assessment in patients with glioma. Methods The 62 astrocytoma samples of various pathological gradings and 11 samples of normal cerebral tissues were subjected to SABC immunohistochemistry assay and reverse transcription polymerase chain reaction (RT-PCR) for determination of RhoC protein and mRNA expression. Results In contrast to weak or absence of expression, RhoC protein tested positive in 62 glioma tissues, as suggested by immunohistochemistry assay. Astrocytoma of various grades exhibited difference in the proportion of cells with positive RhoC expression and the ratio of RhoC mR -NA and GAPDH mRNA gray scale. No difference was revealed between grade I and grade II as well as between grade M and grade IV (all P<0.05). Higher RhoC mRNA expression was found in grade M and IV as compared with grade I and II. Additionally, the proportion of RhoC positive cells was positively correlated with RhoC mRNA expression and pathological grading of glioma (r=0.814 and 0.863, respectively, both P<0.01). Conclusion RhoC expression is present in all 62 glioma samples, the extent of which is positively correlated with pathological grading, sug -gesting that RhoC may serve as a novel and efficient diagnostic marker for grade I to IV astrocytoma and a complement to routine criteria for histopathological grading.
