目的:研究P53基因状况与抗癌化疗敏感性的关系,对抗癌药耐药机制进行探讨。方法:用MTT法测定17种抗癌药对86例恶性肿瘤的抑制率,并用聚合酶链反应——单链构象多态分析技术检测其P53基因状况。结果:统计比较抗癌药在P53基因正常组和P53基因异常组中的抑制率,发现多数抗癌药抑制率前者显著地大于后者。结论:当恶性肿瘤中存在P53基因突变或缺失时,抗癌药耐药性显著增加。%OBJECTIVE: To testify the relationship between the P53 gene status and chemosensitivity and explore the mechanism of antitumor drug's resistance. METHOD: Using MTT method, we tested the inhibition rates of 17 antitumor drugs in 86 cancer patients, and also analysed the P53 gene status with polymerase chain reaction-single stranded conformation polymorphism in it. RESULTS: The inhibition rates of majority antitumor drugs in the P53 gene normal group were significantly higher than in the P53 gene abnormal group. CONCLUSION: When the deletion and mutation of P53 gene occurred in the malignant tumours, the antitumor drugs' resistance increased significantly.
