实时荧光定量PCR检测WISP-1/NGX6基因体系与子宫颈癌细胞增殖转移的关系

摘要

Objective To explore the correlation between oncogene WISP-1 and tumor-suppressor gene NGX6 expression and cervical cancer cell proliferation and/or metastasis.Methods Primers and probes were designed and synthesizes based on the mRNA sequence of human WISP-l and NGX6 gene, respectively. Total RNA were extracted from human normal cervixtissues (30 cases) as well as non-metastasis and metastasis group (30 cases for each group) of cervix invasive cancer (cervical squamous cell carcinoma) tissues, and the expressions of WISP-l and NGX6 gene were detected and compared by real-time quantitative fluorescence PCR.Results The expression of the oncogene WISP-l in both metastasis and non-metastasis group of cervical cancer tissues were significantly higher than normal cervical tissues (with P < 0.001 for both). Moreover, WISP-l gene expression in the metastasis group was significantly higher when compared to the non-metastasis group of cervical cancer tissues (P < 0.001). The expression of tumor-suppressor gene NGX6 in both metastasis group and non-metastasis group of cervical cancer tissues were lower than normal cervical tissues (with P < 0.001 for both). Similarly, NGX6 gene expression in the metastasis group was significantly lower when compared to the non-metastasis group of cervical cancer tissues (P < 0.05).Conclusion WISP-1/NGX6 gene expression has correlation with cervical cancer cells proliferation and metastasis.%目的 探讨原癌基因WISP-1和抑癌基因NGX6与子宫颈癌细胞增殖转移的关系.方法 参考人WISP-1、NGX6基因mRNA序列分别设计引物与探针.提取人正常子宫颈组织(30例)、子宫颈浸润癌(均为子宫颈鳞状细胞癌)组织未转移组及转移组(各30例)总RNA,采用实时荧光定量PCR技术分别检测各组标本组织中WISP-1、NGX6基因的表达量并进行对比分析.结果 原癌基因WISP-1在子宫颈癌组织转移组及未转移组中的表达量均明显高于正常子宫颈组织(均P＜ 0.001),在子宫颈癌组织转移组中的表达量明显高于未转移组(P＜0.001).抑癌基因NGX6在子宫颈癌组织转移组及未转移组中的表达量均低于正常子宫颈组织(均P＜ 0.001),在子宫颈癌组织转移组中的表达量明显低于未转移组(P＜0.05).结论 WISP-1/NGX6基因体系与子宫颈癌的细胞增殖有关,并且与子宫颈癌细胞的转移亦有相关性.
