CiaRH生物信息学分析及其在不同猪链球菌血清型中的分布

         

摘要

目的 对猪链球菌2型中二元信号转导系统CiaRH的序列进行分析并评估该系统在不同血清型中的分布.方法 利用生物信息学方法分析其组分蛋白的结构域.PCR及序列分析手段评估CiaRH在不同血清型猪链球菌中的分布情况.结果 比对结果显示,CiaRH同源蛋白存在于链球菌属的不同种细菌中.结构分析发现,在CiaH的N-端包含两个跨膜区,其间的间隔区域发现一个可能用来结合环境信号的PDC结构域;CiaH的C-端为保守的HisKA及HATPase_C结构域.PCR及序列比对结果显示,CiaRH在不同血清型猪链球菌中广泛分布.结论 提示CiaH为胞外感应型组氨酸激酶.在CiaR的N-端为保守的信号接收结构域REC,而C-端为具有DNA结合功能的trans_reg_C结构域.%This study aimed to analyze the sequences of a two-component signal transduction system (TCSTS) CiaRH from S. suis 2 and evaluate the distribution of this TCSTS on different serotypes . The BLAST results showed that the CiaRH orthologs were found in different species of the genus Streptococcus. Structure analysis on CiaRH revealed that two transmem-brane regions existed at the N-terminal of CiaH , between which a PDC domain that would anticipate in signals binding was predicted ; the highly conserved domains HisKA and HATPase_C were found at the C-terminal of the protein . These results suggest that the CiaH belongs to the periplasmic (or extracellular )-sensing HKs group . The conserved REC domain that involved in signal receiving and the DNA binding domain trans_reg_C were found located at the N-terminal and C-terminal of CiaR , respectively . Combined results of PCR detection and BLAST sequence suggest that the CiaRH are widely distributed in different serotypes of S. suis.
