海南不同产地草豆蔻的差异蛋白分析

         

摘要

As a typical plant that had the homology of medicine and food,A lpinia Katsumadai had been studied on its main chemical components.But there was few research about vegetable proteins.The protein information of Alpinia Katsumadai in Hainan Baisha and Bawangling were investigated in this paper.The protein profiles of Hainan A lpinia Katsumadai were obtained by 2-dimensional polyacryamide gel electrophoresis (2-DE) technique.The high abundance proteins in different habitats were compared and screened out by software Image Master5.0.The protein database was searched by Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry (MALDI-TOF MS) and the software Mascot Distiller.The results indicated that there were 40 high abundance proteins of Alpinia Katsumadai in two areas.The identical proteins were removed and the identified proteins were assigned to 22 kinds of proteins which related to the antimicrobial activity of plant,stress response,cellular differentiation and proliferation,physical characteristics.Some differential proteins were found to have certain antimicrobial activity.They were antimicrobial peptides of 11,12 and 13 protein spots,cell division protein FtsZ of 7 and 25 protein spots,E3 ubiquitin-protein ligase of 24 protein spot,17.7 kDa heat shock protein of 31 protein spot and glutathione S-transferase of 14 and 35 protein spots.These differential proteins were related with the anti-inflammatory and bacteriostasis and low toxicity of Alpinia Katsumadai.This paper filled the information of biomolecules of Alpinia Katsumadai and illustrated the biological diversity features of Alpinia Katsumadai in different habitats.The study may provide scientific basis for further development and utilization of A lpinia Katsumadai and fully understand its pharmacological activities.%草豆蔻为典型的药食同源植物,关于其主要的化学成分已早有研究,但对其含有的植物蛋白却鲜有研究.本研究首次探索了海南地道草豆蔻(白沙和霸王岭2个产地)中所含蛋白信息.应用双向电泳进行蛋白分离得到相应全蛋白图谱,并用Image Master5.0软件比较筛选出不同产地的高丰度差异蛋白,通过基质辅助激光解析-电离飞行时间质谱分析(Matrix Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrum,MALDI-TOF MS)及Mascot Distiller软件对蛋白进行搜库.结果显示:2个不同产地草豆蔻共有40个高丰度差异蛋白.去除相同的蛋白,经分析确定这些差异蛋白归属22种植物蛋白,主要涉及植物的抗菌活性、应激反应、细胞分裂增殖及对植物物理性状的影响等方面.首次发现某些差异蛋白可能具有一定的抗菌活性,如11、12及13号的类似豌豆的抗菌肽段,7或25号的纤维状细胞分裂蛋白,24号的泛素蛋白连接酶E3,31号的小分子热休克蛋白,14及35号的谷胱甘肽s-转移酶.这些差异蛋白均与草豆蔻极其主要的活性组份所具有的抗炎抑菌、毒性低等的特点相关.本研究结果不仅是对草豆蔻所含物质中的生物大分子种类的补充和完善,同时也体现了不同产地草豆蔻的生物多样性特点,为深入利用、研究这种药食同源的植物及全面了解其药理活性提供了一定的科学依据.
