基于中医四诊方法对哮喘大鼠肾阳虚证模型建立的评价研究

摘要

目的:建立并评价哮喘大鼠肾阳虚病证结合模型.方法:取雄性SD大鼠30只,随机分为正常组、哮喘模型组、肾阳虚哮喘模型组,每组10只.肾阳虚哮喘模型组采用地塞米松混悬液灌胃结合卵蛋白雾化激发的方法,建立肾阳虚证哮喘病证结合模型.哮喘模型组采用直接卵蛋白雾化激发的方法建立哮喘模型.正常组以生理盐水替代致敏及雾化.全过程观察其形态、行为学等变化,并定时测量其体质量、爪温、腋温、爪尾r值等数据.结果:哮喘模型组及肾阳虚哮喘模型组大鼠均有明显咳嗽、喷嚏、烦躁等现象出现.肾阳虚哮喘组大鼠爪尾颜色肉眼观变淡,触其皮肤明显变凉,精神萎靡、一般状态较差;爪温、腋温均降低,低于正常组及哮喘模型组(P＜0.05);造模过程中肾阳虚哮喘组大鼠体质量增长速度明显变缓,体质量小于正常组及哮喘模型组(P＜0.05),停用地塞米松后体质量仍明显较轻;肾阳虚哮喘模型组爪尾r值均明显低于哮喘模型组及正常组(P＜0.05).结论:本实验运用地塞米松灌胃复合卵蛋白雾化激发哮喘的方法建立哮喘肾阳虚证病证结合模型.从大鼠四诊信息对哮喘动物肾阳虚证候评价,最终认为该模型建立成功.%Objective:To establish and evaluate the asthma model with kidney-yang deficiency.Methods:30 male SD rats were taken and randomly divided into 3 groups,normal control group,asthma model group,asthma model group with kidney-yang deficiency.Asthma model group of kidney-yang deficiency was given intragastric administration by dexamethasone suspension and induce asthma by 2％ ovalbumin inhalation.Asthma model group was induced asthma by 2％ ovalbumin inhalation.The normal control group was replaced for normal saline to sensitize and induce asthma.To observe the changes of general condition and measure temperature of axillaries and paws of all rats and chroma values of the RGB.Results:The general condition of asthma model group of kidney-yang deficiency was the worst,rats showed cough and sneezing.The weight of the normal control group and asthma group was obviously higher than that in asthma model group of kidney-yang deficiency (P ＜ 0.05).The temperature of the axillaries and paws in the normal control group were higher than that in other group(P ＜ 0.05).R values of the paw and tail in asthma model group of kidney yang deficiency were lower than those in the normal control group and asthma group (P ＜ 0.05).Conclusion:The method which was combined intragastric administration by dexamethasone suspension and ovalbumin inhalation to duplicate the asthma combination model of disease and syndrome of the-kidney Yang deficiency was successful.
