交通任督针法对血管性痴呆大鼠认知能力及海马组织5-HT含量的影响

         

摘要

目的:探讨交通任督针法对血管性痴呆(VD)大鼠认知能力及海马组织5-羟色胺(5-HT)含量的影响,为临床治疗血管性痴呆提供依据.方法:将40只SD雄性大鼠随机分为4组,即:正常组、模型组、交通任督法针刺组、西药对照组.采用双侧颈动脉夹闭再通法制备VD大鼠模型.造模成功后,进行治疗;治疗结束后用Morris水迷宫测试认知能力,然后取大鼠海马组织,采用酶联免疫吸附法检测大鼠海马组织单胺类神经递质5-羟色胺(5-HT)的含量.结果:Morris水迷宫结果显示,定位航行试验正常组大鼠平均逃避潜伏期短于模型组(P<0.05);交通任督法针刺组、西药组明显短于模型组(P<0.05);空间探索试验大鼠平均跨越平台次数模型组低于正常组(P<0.05),交通任督法针刺组、西药组大鼠平均跨越平台次多于模型组(P<0.05).海马组织5-HT含量检测结果显示,模型组海马组织5-HT含量低于正常组(P<0.05),交通任督法针刺组、西药组5-HT含量高于模型组(P<0.05).结论:交通任督法针刺对于VD大鼠认知能力有一定的改善,提高海马组织5-HT的含量是其作用机制之一.%Objective:To provide basis for the treatment of vascular dementia (VD) in clinic by observing the effects of Jiaotong Rendu Acupuncture on cognitive ability and 5-HT content in hippocampus of rats.Methods:40 SD male rats were randomly divided into four groups,namely:the normal group,model group,Jiaotong Rendu Acupuncture group and western medicine group.VD models were made by bilateral carotid artery clip recanalization method.Ater successful modeling,rats were given the corresponding treatment;after the treatment,Morris water maze test was used to determine rats' cognitive abilities,using enzyme-linked immunosorbent (ELISA) method to detect 5-HT content in hippocampus of rats.Results:Morris water maze results show that the navigation test average escape latency of rats in normal group was shorter than that of the model group (P < 0.05);the average escape latency of acupuncture group and western medicine was shorter than that in model group (P < 0.05);space exploration test the average number of cross platform of model group rats was fewer than that of the normal group (P < 0.05),the average number of cross platform of acupuncture group and western medicine group was more than that of model group (P < 0.05).The result of 5-HT content in hippocampus determination showed that 5-HT content in hippocampus of model group was significantly lower than that of the normal group (P < 0.05),5-HT content in acupuncture group and western medicine group was more than that of model group (P < 0.05).Conclusion:Up-regulating 5-HT content in hippocampus was one of the mechanism of improvement effect of Jiaotong Rendu Acupuncture on cognitive ability of VD rats.
