艾灸大椎穴对慢性应激大鼠神经营养因子的影响

     

摘要

AIM:To observe the effect of moxibustion at Dazhui point on hippocampus neurons and the expression of brain-derived neurotrophic factor(BDNF) of these cells in the chronic mild stress rats. METHODS:Eighteen health male SD rats were randomly divided into control group,model group and moxibustion group.The chronic stress model was prepared by individual breeding and a variety of unforeseeable chronic stress.Under light microscope,the morphological changes of hippocampus neurons were detected by hematoxyli-eosin(HE) staining,and the expression of BDNF was detected by immunohistochemistry. RESULTS:The neurons could be injured by the chronic mild stress.And the decrease of BDNF positive neurons was detected in the model rats.The moxibustion at Dazhui point could protect the neurons injured by chronic mild stress.In moxibustion group the means of optical density of neurons were(16.98± 1.34)% (CA2), (32.77± 2.38)% (CA3),( 2.48± 0.25)% (CA4).And in the model group the means of optical density of neurons were(12.21± 1.58)% (CA2), (19.89± 3.82)% (CA3),( 1.04± 0.38)% (CA4)(F=6.25,P< 0.05;F=12.27,11.45,P< 0.01). CONCLUSION: The Moxibustion at Dazhui point effectively regulates the expression of BDNF and protects hippocampus neurons in chronic mild stress rats.%目的:试图发现艾灸大椎穴对慢性应激状态下大鼠海马神经元及脑源性神经营养因子( brain-erived neurotrophic factor,BDNF)的影响. 方法 :SD雄性大鼠 18只,按随机数字表法分为正常组、模型组、艾灸组.应用孤养和长期不可预见性的中等强度刺激造成慢性应激失调模型,观察各组大鼠海马神经元的变化.采用苏木精-伊红( HE)染色、尼氏体染色的方法光镜下观察海马神经元形态的改变,用免疫组织化学的方法对海马 BDNF阳性神经元进行染色,并用图像分析仪定量分析. 结果:慢性应激可致大鼠海马神经元明显受损, BDNF阳性神经元数量亦显著减少,形态以空泡为主.与模型组比较,艾灸穴对此有明显改善作用.艾灸组海马 BDNF阳性神经元的平均光密度 CA2, CA3, CA3区面积百分比: (16.98± 1.34)%, (32.77± 2.38)%,( 2.48± 0.25)%;模型组相应指标分别为 (12.21± 1.58)%, (19.89± 3.82)%,( 1.04± 0.38)%( F=6.25,P< 0.05;F=12.27,11.45,P< 0.01). 结论:艾灸大椎穴对慢性应激大鼠 BDNF有良性调节作用,并对海马神经元有保护作用.
