血清铜和血清铜蓝蛋白活力测定对神经管畸形孕鼠诊断价值的研究

摘要

目的 通过检测神经管畸形模型孕鼠血清铜和血清铜蓝蛋白氧化酶活力的变化,探讨血清铜和血清铜蓝蛋白(serum ceruloplasmin,sCP)是否可以预测神经管畸形胚胎的早期发生.方法 雌性Wistar孕鼠按随机数字表法分组:脊柱裂模型组在孕鼠第10天给予维甲酸致畸,建立神经管畸形胎鼠模型,正常对照组给予同等剂量的橄榄油.于孕11、12、14、16、18、20 d在雌鼠心尖部取血,分离血清,采用联大茴香胺酶比色法测定sCP的酶活力,采用络合比色法测定血清铜离子含量.结果 经维甲酸致畸后,孕11d脊柱裂模型组sCP酶活力有升高趋势但差异无统计学意义(P>0.05),而铜离子含量有显著升高(P<0.05),孕12、14、16d脊柱裂模型组sCP活力和铜离子含量均明显高于正常对照组(P<0.05),而孕18、20 d脊柱裂模型组sCP活力和铜离子含量与正常对照组比较差异无统计学意义(P>0.05).结论 sCP酶活性和血清铜离子含量在神经管畸形孕鼠明显增高,测定sCP酶活性和血清铜离子含量对胚胎神经管畸形早期诊断存在一定价值.%Objective To explore the changes of serum copper and oxidasic activity of serum ceruloplasmin (sCP) in pregnant rat model with neural tube defects for biomarker screening of prenatal diagnosis.Methods The pregnant rats were randomly divided into two groups.Spina bifida aperta was induced with a single intragastric injection of atRA at E10.Animal in normal control group was injected with the same amount of atRA-free olive oil on the same day.Blood samples from pregnant rats were obtained via cardiac puncture at E11,E12,E14,E16,E18 and E20.The level of serum copper and oxidasic activity of sCP were tested respectively by complexation colorimetric determination and O-dianisidine dilydrochloride catalyzed by sCP colorimetric determination.Results In atRA-fed group,the oxidasic activity of sCP had a rising trend at E11 and yet there was no significant differences (P>0.05).The serum copper content increased significantly at the same gestation day (P<0.05).Both oxidasic activity of sCP and serum copper content in atRA-fed group were obviously higher than those in control group from gestation day 12 to 16 (P<0.5).However,oxidasic activity of sCP and serum copper content in atRA-fed group showed no significant difference as compared with control group (P>0.05).Conclusions Oxidasic activity of sCP and serum copper content rise obviously in pregnant rats with neural tube defect fetuses.It is helpful to diagnose early development of fetal neural tube defects by detecting the oxidasic activity of sCP and serum copper content.
