藏药萝蒂对视网膜血管保护作用的实验研究

         

摘要

Objective To observe the LuoDi drug on diabetic rats retinal microvascular and the effect of vascular endothelial growth factor .Methods To made in diabetes animal model in SD rats by intraperitoneal injection of STZ ,40 rats that successful model were randomly divided into four groups ,the negative control group ,Luodi's high ,medium and low dose group ,at the same time normal control group ,10 SD rats was normal group ,after gavage march observed between groups of rat retinal capillary morphology , detection of retinal capillary basement membrane thickness ,to detect the contents of vitreous VEGF .Results The negative control group rats retinal capillary expansion ,markedly thickened basal membrane , proliferation of endothelial cells obviously ,the shape is irregular ,perithelial cell showed a trend of decrease . High dose treatment group rats retinal capillary basement membrane thickening improved ,endothelial cell morphological rules .Luodi's treatment group after treatment basement membrane thickness thinning to different extent ,Luodi's middle and high dose group of capillary basement membrane thickness before and after treatment comparison difference was statistically significant .Luodi's middle ,high dose group of vitreous VEGF content is significantly lower than the negative control group of rats ,have significant difference (P<0 .01) .Conclusion Luodi drug has certain significant protective effect on STZ diabetic rats retinal capillary morphology and retinal capillary basement membrane thickness ,it can reduce vitreous VEGF .%目的:观察萝蒂对糖尿病大鼠视网膜微血管及血管内皮生长因子的影响;方法 STZ腹腔注射SD大鼠造模糖尿病大鼠动物模型,造模成功40只大鼠,随机分为4组,即阴性对照组、萝蒂高、中、低剂量组,同时10只SD大鼠设正常对照组,灌胃3月后观察各组大鼠视网膜毛细血管形态学,检测视网膜毛细血管基底膜厚度,检测玻璃体V EG F含量。结果阴性对照组大鼠视网膜毛细血管明显扩张,基底膜显著增厚,内皮细胞明显增生,形态不规则,周细胞呈减少趋势。萝蒂高剂量治疗组大鼠视网膜毛细血管基底膜增厚有所改善,内皮细胞形态规则。萝蒂治疗各组治疗后基底膜厚度有不同程度变薄,萝蒂中、高剂量组毛细血管基底膜厚度治疗前后比较差异有统计学意义。萝蒂中、高剂量组大鼠玻璃体V EG F含量明显低于阴性对照组,差异有显著意义(P<0.01);结论萝蒂对STZ糖尿病大鼠视网膜毛细血管形态学及视网膜毛细血管基底膜厚度均有明显的保护作用,能够降低玻璃体VEGF。
