VEGF受体3在人脑胶质母细胞瘤中的表达及作用

         

摘要

目的 研究血管内皮细胞生长因子受体3在人脑胶质母细胞瘤中的内皮细胞中的表达,其对肿瘤血管内皮细胞增殖,迁徙和血管成形功能的作用.方法 使用实时聚合酶链式反应(RT-PCR),酶联免疫吸附法(ELISA)研究血管内皮细胞生长因子C在脑胶质瘤组织和肿瘤细胞U87、U251中的表达,使用RT-PCR,免疫组化方法研究血管内皮细胞生长因子3在脑胶质母细胞组织和内皮细胞中的表达,使用特异性阻滞剂MAZ51阻滞其在人脑胶质母细胞瘤中的内皮细胞中的作用.结果 血管内皮细胞生长因子C表达于人脑胶质瘤组织和肿瘤细胞中,血管内皮细胞生长因子3表达于人脑胶质母细胞瘤组织和内皮细胞中,使用阻滞剂MAZ51可降低肿瘤血管内皮细胞增殖,迁徙和血管成形能力.结论 血管内皮细胞生长因子受体3参与人脑胶质母细胞瘤血管拟态形成,可能成为脑胶质瘤肿瘤抗血管治疗的新靶点.%Objective The expression of vascular endothelial growth factor receptor 3 (VEGFR-3) and its function in the process of proliferation,migration and tube-formation of human glioblastoma endothelial cells were explored.Methods The expression of vascular endothelial growth factor-C (VEGF-C) in human glioblastoma tissues and cell lines were detected by real-time polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA),while the expression of VEGFR-3 in human glioblastoma tissues and endothelial cells was studied by RT-PCR and immunohistochemistry.The effect of VEGFR-3 on the proliferation,migration and angioplasty of tumor vascular endothelial cells was investigated by blocking VEGFR-3 with special blocker MAZ51.Results VEGF-C could be found on glioblastoma tissues and cell lines.VEGFR-3 was expressed on glioblastoma tissues and endothelial cells,inhibition of VEGFR-3 could influence the proliferation,migration and tube formation ability of tumor vascular endothelial cells.Conclusion VEGFR-3 is involved in mimicry of human glioblastoma angiogenesis,which can be a new target of anti-angiogenesis therapy in human glioblastoma.
