DNA-PKcs蛋白表达对鼻咽癌预后影响的研究#

摘要

Objective:To investigate the expressions of DNA-dependent protein kinase catalytic subunit(DNA-PKcs) in nasopharyngeal cancinoma and their relationships with clinical outcome.Methods:Between May 2007 and February 2012,all 100 untreated patients with NPC,who were planned to receive IMRT at the People’s Hospital of Guangxi Autonomous Region were enrolled in this study.However,13 of 100 patients lost to follow-up.The expression of DNA-PKcs proteins in the tissue of nasopharyngeal carcinoma was detected by immunohistochemistry and then combined with all the clinical data to analyze the relationship between the expression of the proteins and prognosis of the patients.Results:The overexpression of DNA-PKcs was 60%and low expression 40%in 87 cases of patients. No significant differences were found between the expression of the proteins and short-term effect(P=0.348).Howerer,There was a significant positive correlation between the expression level of DNA-PKcsand survival time of patients(r=0.234,P=0.048). Moreover,patients with high DNA-PKcs expression had significantly better distant metastasis-free survival than patients with low levels of DNA-PKcs(P=0.001).Conclusion:There was no significant correlation between the expression level of DNA-PKcs and short-term effect. Lower expression of DNA-PKcs might have higher distant metastasis in patients with NPC.%目的：研究DNA-PKcs蛋白表达与鼻咽癌预后的关系。方法：收集2007年5月~2012年2月经广西壮族自治区人民医院病理学确诊为鼻咽癌且选择调强放疗的患者100例。其中13例失访。采用免疫组化SP法检测87例患者中DNA-PKcs蛋白在鼻咽癌组织中的表达,分析DNA-PKcs蛋白表达与患者近期疗效、总生存以及发生远处转移之间的关系。结果：87例患者DNA-PKcs高表达60.0%,低表达40%。DNA-PKcs的表达水平与患者的近期疗效无关(P=0.348)；但与患者生存时间呈正相关(r=0.234,P=0.048),单因素分析显示DNA-PKcs低表达者较高表达者更容易发生远处转移(P=0.001),而且生存时间更短(P=0.007)。结论：鼻咽癌组织中DNA-PKcs的表达水平与患者近期疗效无关,DNA-PKcs低表达可能作为预测鼻咽癌患者预后较差的一个指标。
