神经生长因子对哮喘小鼠TNF-α表达的影响

         

摘要

Objective To explore the influence of NGF on the expression of TNF-α in asthmatic mice .Methods A mouse mod-el of asthma was induced by sensitization and airway challenged with ovalbumin .30 BALB/c mice were divided into 3 groups :the normal control group ,the asthma group ,and the NGF-blocking group .The TNF-α protein was detected by ELBA and the messenger RNA (mRNA) of TNF-α in the lung was tested by reverse transcription polymerase chain reaction (KF-PCR) .Results The levels of TNF-α(pg/ml)in serum of the three groups were 114.61 ± 18 .28,196 .83 ± 46 .05,136 .27 ± 24.68 .The TNF-α level in serum of the asthma group was higher than that of the normal control group and the TNF-α level in serum of the NGF-blocking group was lower than that of the asthma group(ρ<0 .05) .Compared to the normal control group ,the TNF-α mRNA level was elevated in the asthmatic mice ,and the TNP-α mRNA level was lower in NGF-blocking group than that in the asthma group ( ρ<0 .05) .Conclusion TNP-α participates in the development of asthma .NGF may be involved in the pathogenesis of asthma by enhancing the express of TNF-α.%目的 探讨神经生长因子(NGF)对哮喘小鼠TNF-α表达的影响.方法 卵白蛋白致敏并雾化吸入激发建立小鼠哮喘模型.30只小鼠随机分为3组:正常对照组、哮喘组、NGF阻断组.ELISA法测定血清TNF-α的水平,RT-PCR法检测肺组织TNF-α mRNA的表达.结果 各组血清TNF-α水平(pg/ml)分别为114.61±18.28、196.83±46.05、136.27±24.68,哮喘组血清TNF-α含量显著高于正常对照组,NGF阻断组血清TNF-α含量低于哮喘组(P<0.05).哮喘组较正常对照组肺组织TNF-α mRNA表达明显增强,NGF阻断组肺组织TNF-α mRNA表达较哮喘组减弱(P<0.05).结论 TNF-α参与哮喘发病,NGF可能通过上调TNF-α的表达参与哮喘的发病.
