壳聚糖纳米粒子基因载体的制备与相关研究

         

摘要

Objective To prepare liquid of CS/pcDNA3. 1/ GDNF and examine the related properties of chitosan as gene vector. Methods To make liquid of CS/pcDNA3. 1/ GDNF using recondensing method and observe physical prop-erties,stability,gene linking rate,gene protecting function,slowly releasing effect,gene loading ability and gene loading rate. Results The size of chitosan is at the range of 106-228 nm,thc properties of chitosan is stable,ehitosan has good gene carrying ability and can protect gene it carried whose gene loading ability and gene loading rate is high. Conclusion chitosan made by recondensing method can satisfy the need of gene vector,it has been proved to be an ideal gene vector.%目的 制备CS/pcDNA3.1/GDNF混悬液,并对壳聚糖作为基因载体的相关性能进行检测.方法 采用复凝聚法制备CS/pcDNA3.1/GDNF混悬液,观察壳聚糖纳米粒子的物理性能,稳定性,基因结合效率,基因保护功能,缓慢释放效果及基因负载力和负载率.结果 壳聚糖大小为106-228 nm,性质稳定,具有良好的基因携带功能,能够有效地保护所携带基因,基因负载力及基因负载率较高.结论 应用复凝聚法制得壳聚糖纳米粒子,能够满足基因载体的需要,是一个较理想的基因载体.
