肾炎防衰液对糖尿病肾病大鼠的防治作用及对于 MCP -1表达的影响

         

摘要

Objective:To study the effect of the Shenyanfangshuai Solution(the Traditional Chinese Medical Solution for preventing and treating renal failure)on the therapy of diabetic nephropathy(DN)and the expression of MCP - 1 for STZ - induced rats. Methods:The model rats with the diabetic nephropathy(DN)were treated by the nephrectomy and intraperltoneal injection of streptozotocin. All model rats were randomly divided into model group,benazepril group and Shenyanfangshuai Solution group. The healthy rats were chosen as sham - operation group. All rats were given the relevant solutions respectively for 8 weeks. The rats of benazepril group were taken by intragastric administration with the benazepril suspension liquid(1. 0 mg·kg - 1 ·d - 1 ),and the rats of Shenyanfangshuai Solution group intragastric administration with the Shenyanfangshuai Solution(11. 4 g·kg - 1 ·d - 1 )respective-ly. The rats of model group and sham - operation group were taken by intragastric administration with physiological saline. When the trial was finished,the serum creatinine and urea nitrogen were detected. Renal pathology was detected by HE and Masson staining. The expression of MCP - 1 in renal tissue was detected by immnuohistochemistry. Results:There was no significant difference about serum urea nitrogen level in each group of rats(P ﹥ 0. 05). The values of serum creatinine in model group were significantly higher than that of the sham - operation group(P ﹤ 0. 05). Compared with model group,the values of serum creatinine in Shenyanfangshuai Solution group’s were decreased significantly(P ﹤ 0. 05). Glomerularsclerosis index,renal tubule injury index,renal tubular inter-stitial fibrosis index and the expression of MCP - 1 in renal tissue of model group were significantly higher than that in sham - opera-tion group(P ﹤ 0. 05). Compared with model group,the levels of the three above - mentioned indices and the expression of MCP - 1 in Shenyanfangshuai Solution group’s were decreased significantly(P ﹤ 0. 05). Conclusion:Shenyanfangshuai solution can decrease the value of serum creatinine,and the extent of glomerulosclerosis and renal interstitial fibrosis,and it has the function of down - regulating the expression of MCP - 1 in DN rats. So Shenyanfangshuai solution can prevent the progress of diabetic nephropa-thy in DN rats.%目的:研究肾炎防衰液对糖尿病肾病大鼠的防治作用。方法:建立大鼠糖尿病肾病模型,随机分为模型组、贝那普利组和肾炎防衰液组,另设假手术组;给予贝那普利组和肾炎防衰液组分别灌胃以贝那普利混悬液(1.0 mg·kg -1·d -1)和肾炎防衰液(11.4 g·kg -1·d -1);模型组和假手术组灌以等量生理盐水。实验进入第8周后取材,检测血清肌酐、尿素氮,采用 HE 和 Masson 染色检测肾脏病理、免疫组化法检测肾组织 MCP -1的表达。结果:各组大鼠血清尿素氮水平差异无统计学意义(P ﹥0.05);模型组血清肌酐水平明显高于假手术组(P ﹤0.05),而肾炎防衰液组较模型组为低(P ﹤0.05);模型组肾小球硬化指数、肾小管损伤指数、肾小管间质纤维化指数以及肾组织中 MCP -1表达显著高于假手术组(P ﹤0.05);肾炎防衰液组肾小球硬化指数、肾小管损伤指数、肾小管间质纤维化指数以及 MCP -1表达均较模型组为低(P ﹤0.05)。结论:肾炎防衰液可以降低血清肌酐水平,减轻糖尿病肾病大鼠肾小球硬化及肾间质纤维化的程度,下调糖尿病肾病大鼠肾组织 MCP -1的表达,延缓糖尿病肾病进展。
