通心络下调基质金属蛋白酶稳定动脉粥样硬化斑块的作用研究

         

摘要

Objective Through observing the effect of Tongxinluo on regulating matrix metal proteinase (MMP-2/9) levels and stabilizing the atherosclerosis (AS) plaques to discuss the effective mechanism of Tongxinluo for treatment of acute coronary syndrome (ACS). Methods Thirty-eight New Zealand rabbits were randomly divided into five groups: normal (A group, 6 cases), model (B group, 8 cases). Tongxinluo treated (C group, 8 cases), simvastatin treated (D group, 8 cases) and Tongxinluo plus simvastatin treated (E group, 8 cases) groups. A ball pouch was applied to damage abdomen aorta and a high fat diet was used to establish rabbit AS model. The rabbits in medicine treated groups were given respective medicine and the blood lipid was detected 8 weeks later; simultaneously the blood stream velocities of iliac artery and aorta were examined with Doppler apparatus and MMP-2/9 levels were detected through immunohistochemistry. Results MMP-2/9 expressions and blood lipid level in B group were higher than those in A group ; compared with B group, MMP-2/9 positive expressions were significantly lower in C, D and E groups, being the lowest in E group (MMP-2: 25 651. 7± 2 185.8 vs. 846 907. 8± 32 285.6, MMP-9: 24 258.1±1 499.5 vs. 942 219. 5±21 857.9, all P<0. 05); total cholesterol (TC, mmol/L), triglyceride (TG, mmol/L) and low density lipoprotein (LDL, mmol/L) in serum all were decreased in certain degrees of level in C, D and E groups, being reduced the most in E group (TC: 32. 51±0. 88 vs. 38. 56±1. 91, TG: 7.15±1. 87 vs. 8. 07±l. 68, LDL: 25. 47±3. 56 vs. 32. 58±2. 14, all P<0. 05); compared to A group, the velocities of the blood stream were all decreased in B, C, D and E groups, being reduced the most in E group (m/s:21. 55 ±0.78 vs. 35. 95 ±1.34, P<0. 05). Conclusion Tongxinluo can speed up the blood flow velocity and reduce the AS plaque formation, which may be related to the stabilization of plaques through decreasing MMP-2/9 and associated factor expressions.%目的 通过观察通心络调节基质金属蛋白酶-2/9(MMP-2/9)的活性及稳定动脉粥样硬化(AS)斑块的效应,探讨通心络治疗急性冠脉综合征(ACS)的有效机制.方法 应用球囊损伤腹主动脉加高脂饮食建立兔AS模型.将38只新西兰兔按数据列表法分为手术对照组(A组,6只),AS模型组(B组,8只)、通心络治疗组(C组,8只)、辛伐他汀治疗组(D组,8只)和通心络+辛伐他汀治疗组(E组,8只).给药8周后检测血脂,用多普勒检测腹主动脉血流速度;用免疫组化染色检测MMP-2/9的表达.结果 B组MMP-2/9表达、血脂水平较A组明显升高;与B组比较,C,D,E组MMP-2/9阳性表达(A值)明显减少,以E组表达水平最低(MMP-2:25 651.7±2 185.8比846 907.8±32 285.6,MMP-9:24 258.1±1 499.5比942 219.5±21 857.9,均P<0.05);C,D,E组血清总胆固醇(TC,mmol/L)、三酰甘油(TG,mmol/L)、低密度脂蛋白(LDL,mmol/L)均有不同程度降低,以E组降低最显著(TC:32.51±0.88比38.56±1.91,TG:7.15±1.87比8.07±1.68,LDL:25.47±3.56比32.58±2.14,均P<0.05);B,C,D,E组较A组腹主动脉血流速度下降,以E组下降趋势最缓慢(m/s:21.55±0.78比35.95±1.34,P<0.05).结论 通心络有加快血流速度及减少AS斑块形成的作用,其机制可能与下调MMP-2/9及其相关因子表达、稳定斑块有关.
