丹参多酚酸预处理及后处理对大鼠心肌线粒体呼吸链的影响

         

摘要

目的 探讨丹参多酚酸预处理及后处理对大鼠离体心肌缺血/再灌注(I/R)损伤的保护作用.方法 应用紫外分光光度计检测对照组(n=8)、I/R组(n=8,全心缺血30 min、再灌注60 min)和丹参多酚酸预处理组、后处理组(各组n=8,I/R前、后灌注丹参多酚酸10 mmol/L)大鼠心肌线粒体呼吸链酶复合体Ⅰ~Ⅳ活性的变化.结果 I/R组心肌线粒体呼吸链酶复合体Ⅰ~Ⅳ活性(μmol·mg-1·min-1)明显低于对照组(复合体Ⅰ:4.49±0.30 比22.94±2.14,复合体Ⅱ:5.34±0.81比15.80±1.07,复合体Ⅲ:5.40±0.54 比15.67±2.90,复合体Ⅳ:10.19±2.29 比26.08±2.23,均P<0.05).丹参多酚酸预处理及后处理组复合体Ⅰ~Ⅳ活性明显高于I/R组,且丹参多酚酸预处理组较后处理组心肌线粒体呼吸链酶复合体Ⅰ~Ⅳ活性明显升高(复合体Ⅰ:19.92±1.35比16.12±0.43,复合体Ⅱ:9.68±0.88比7.72±0.55,复合体Ⅲ:14.75±0.80比10.37±0.58,复合体Ⅳ:20.59±3.41比14.49±0.86,均P<0.05).结论 丹参多酚酸能够相对增加线粒体呼吸链酶复合体Ⅰ~Ⅳ的活性,改善其能量代谢,减少氧自由基的生成,实现对I/R损伤心肌的保护作用;给予丹参多酚酸预处理作用更优.%Objective To investigate the protective effeci of Salvianolic acid preconditioning and post-conditioning on myocardial ischemia/reperfusion (I/R) injury to isolated rat heart. Methods The changes of activities of myocardial mitochondria! respiratory chain complex Ⅰ-Ⅳ for control group (n = 8), I/R group (n = 8. with ischemia 30 minutes and reperfusion 60 minutes ) and Salvianolic acid preconditioning and post-conditioning groups (n = S, perfusing Salyianolic acid 10 mmol/L before and after I/R respectively ) were determined by ultraviolet spectrophotometry. Results The activities of rnyoeardial rnitoehondrial respiratory chain complex I-IV (μmol·mg-1·mm -1 ) rn I/R group were less than those in control group (complex I ; 4.49 + 0.30 vs. 22.94 + 2.14. complex II ; 5.34+0.81 vs. 15.80 ± 1.07, complex Ⅲ: 5.40 ±0.54 vs. 15.67 ± 2.90, complex Ⅳ : 10.19 ± 2.29 vs. 26.08 ± 2.23, all P<0.05 ) . The activities of rnyoeardial rnitoehondrial respiratory chain complex Ⅰ-Ⅳ in Salvianolic acid preconditioning and post-conditioning groups were obviously higher than those of I/R group, and in preconditioning group, they were significantly higher than those in post-conditioning group (complex Ⅰ; 19.92±1.35 vs. 16.12±0.43, complex Ⅱ ; 9.68 ±0.88 vs. 7.721±0.55, complex Ⅲ; 14.75 ± 0.80 vs. 10.37 ± 0.58, complex Ⅳ: 20.59 ± 3.41 vs. 14.49 ± 0.86. all P<0.05). Conclusions Salvianolic acid has a protective effect on rnyoeardial I/R injury by increasing the activities of myocardial mitochondrial respiratory chain complex Ⅰ-Ⅳ . decreasing the quantity of oxygen free radicals and improving the metabolism of energy. The effect of preconditioning is better.
