小鼠小肠上皮内淋巴细胞的提取与抗原表型测定和功能的初步研究

摘要

Objective:To extract a method for isolating murineintraepithelial lymphocy te and analyze its main function.Methods:Murine intestinal intraepithelial lymphocyte was isolated by a mechanical technique and Percoll d i scontinous density gradient centrigugation.Its surface antigens were measured by using indirect immunoflurescence.Its natural killing activity was examined by L DH method.Results:2×106～4×106iIELs/per mouse were a cquired by using this method,add its vitality＞95%.Of phenotypic antigen,more than 73. 8 % showed CD8+cell,TCRγδ 43.8%.IEL's natural killing activity was high er than PBMCs.Conclusion:This method is effective of isolating IELs.IELs has natural killing activity.%目的：建立一种提取小肠上皮内淋巴细胞的方法，并对其功能进行初步研究。方法：采用机械分离法和Percoll非连续密度梯度离心法提取小鼠小肠上皮内淋巴细胞，经间接荧光染色后用流式细胞仪检测表型。应用乳酸脱氢酶法检测其自然杀伤功能。结果：应用该法可获得小鼠小肠上皮内淋巴细胞2×106～4×106/只鼠，活力测定＞95%。抗原表型中73.8%以上表达CD8分子，TCRγδ为43.8%。而且自然杀伤活性明显高于小鼠脾淋巴细胞。结论：表明采用该法提取小鼠小肠上皮内淋巴细胞有效。IEL具有自然杀伤活性。
