姜黄素联合大豆异黄酮对胰腺癌BxPc-3细胞增殖及凋亡的影响

     

摘要

目的:体外观察姜黄素联合大豆异黄酮对人胰腺癌细胞BxPc-3的增殖和凋亡的影响.方法:采用MTT法观察两者对BxPc-3细胞生长的抑制作用,用Annexin V-FITC/PI双染流式细胞术和caspase-3活性检测法检测细胞凋亡情况.结果:姜黄素和大豆异黄酮单独应用时能抑制BxPc-3细胞的增殖(P<0.01),而两者联合应用后细胞生存率降低更为明显(P<0.01);同样,姜黄素和大豆异黄酮单独作用于BxPc-3细胞时能上调凋亡细胞的比例且升高caspase-3的活性(P<0.05),而联合应用则进一步促进了细胞的凋亡(P<0.05).结论:姜黄素与大豆异黄酮联用对胰腺癌细胞的抑制生长和促凋亡现象有协同作用.%Objective:To investigate the anti-proliferation and apoptotic effect of curcumin combined with soy isoflavone on human pancreatic cancer cell line BxPc-3 in vitro. Methods: The growth inhibition of BxPc-3 cells induced by curcumin and soy isoflavone was determined by MTT assay, and the effect of the two compounds on cell apoptosis were tested with Annexin V-FITC/PI flow cytometry and caspase-3 activity assay. Results: Curcumin and soy isoflavone alone inhibited the proliferation of BxPc-3 cells ( P < 0. 01) , while the cell growth inhibition was significantly greater in the combination of them ( P <0. 01) ; similarly, the apoptotic cell ratio and caspase-3 activity in BxPc-3 cells were up-regulated in response to curcumin or soy isoflavone ( P < 0. 05) , while there was a greater degree of induction of apoptosis in combinatorial treatment group ( P < 0. 05 ). Conclusion: there was synergistic effect of curcumin and soy isoflavone on antiproliferative and apoptotic activity in pancreatic cancer cells.
