槲皮素对Cu2+诱导的β淀粉样蛋白聚集和细胞毒性的影响

摘要

目的 研究槲皮素对Cu2+诱导的β淀粉样蛋白片段(Aβ1～40)聚集和细胞毒性的影响.方法 采用硫磺素T荧光标记研究槲发素对Cu2+诱导的Aβ1～40聚集的影响,利用MTT法测定细胞活力.结果 槲皮素可有效抑制Cu2+诱导的Aβ1～40聚集,对已形成的Cu2+ -Aβ1-40聚集物有明显的解聚作用,能有效防止Cu2+和Aβ1-40共同诱导的SH -SY5Y细胞活力下降.结论 槲皮素对老年痴呆具有潜在的防治作用.%Objective To investigate the effects of quercetin on the aggregation of Ap, _#> induced by Cu2* and on the neurotoxicity of Cu2* -Ap, .^ aggregation. Methods The aggregation of A(3, _w induced by Cu2+ was detected by Thioflavin T fluorescence spectroscopy. The cell viability was evaluated by MTT assay. Results Quercetin could inhibit Cu + -induced aggregation of Ap, _i0 and the preformed Cu2* -Ap, .^ aggregations could be solubilized by quercetin. The neurotoxicity of Cu2* -Ap, _m aggregation to SH-SY5Y cells was attenuated by quercetin and the cell viability loss was significantly decreased. Conclusions Quercetin could be as neuroprotective agent for the treatment of Alzheimer's disease.
