电针对鱼藤酮诱导的帕金森病模型大鼠黑质GDNF mRNA表达的影响

     

摘要

目的 探讨电针对鱼藤酮诱导的帕金森病(PD)模型大鼠黑质GDNF mRNA表达的影响及作用机制.方法 40只大鼠按照随机分组的原则分为空白组、假手术组、模型组、电针组,每组10只.持续4w给模型组和电针组大鼠颈背部皮下注射鱼藤酮(2mg·kg-1·d-1)以诱导其黑质多巴胺能神经元丢失,电针组选取“风府”、“太冲”两穴,频率20 Hz,电压2~4V,连续波,每日1次,每次20 min,7d为1疗程,连续治疗3个疗程;其余各组不行针刺.采用半定量RT-PCR方法检测,观察黑质GDNF mRNA表达变化.结果 长期低剂量颈背部皮下注射鱼藤酮可诱导大鼠出现行为学的改变;模型组大鼠黑质GDNF mRNA表达减少,与空白组、假手术组比较有显著差异(P<0.01);电针组大鼠黑质GDNF mRNA表达增加,与模型组比较有显著差异(P<0.01).结论 电针治疗不仅可以减轻鱼藤酮引起的大鼠行为学异常,而且可以促进PD模型大鼠黑质GDNF mRNA的表达,修复营养DA能神经元,延缓PD的发病进程.%Objective To investigate the influence of electroacupuncture(EA) therapy on the expression of GDNF mRNA of sub-stantia nigra cells in the rotenone-induced Parkinson's disease model rats, explore the mechanism of EA in treatment of Parkinson's disease. Methods Forty rats were averagely and randomly divided into normal, sham-operation, model and EA groups. Highly selective lesions in nigrostriatal dopaminergic neurons in rats were induced by chronic subcutaneous exposure to rotenone at dose of 2 mg ? Kg-1 ? D-1 for 4 w. EA group (20 Hz, 2-4 V, continuous wave) was applied to"Fengfu"and "Taichong"points for 20 min once daily. Other groups were not given treatments. After 21 day treatments, the rats were killed for sampling substantia nigra tissue to detect the expression of GDNF mRNA with semi quantitative reverse transcription polymerase chain reaction (RT-PCR). Results The behavior had obvious changes in rats injected subcutaneously with rotenone. Compared with that of normal group and sham-operation group, the expression of GDNF mRNA in model group was significantly lower( P <0. 01). Compared with that of model group, the expression of GDNF mRNA in EA group was significantly higher( P < 0.01). Conclusions EA therapy not only can change the rotenone-induced abnormal behavior in rats, but improve the expression of GDNF mRNA of substantia nigra cells which can nourish and repair dopaminergic neurons, and delay the process of Parkinson's disease.
