Objective To establish an Alzheimer's disease (AD) cell model induced by okadaic acid (OA) and explore the ultrastructure change of AD cell model.Methods OA was used to induce the AD cell model in SH-SY5Y cell.MTT method was used to measure cell viability.Optical microscope and electron microscopy were used to measure the morphology change of SH-SY5Y cell.Results OA can decrease the viability of SH-SY5Y cell with the time and dose and damage the cell's microtubule.Conclusions OA can induce SH-SY5Y cell to AD cell model.%目的 建立冈田酸诱导的阿尔茨海默病(AD)细胞模型,研究AD细胞模型的超微结构变化.方法 本研究以冈田酸(OA)处理SH-SY5Y细胞建立AD样细胞模型,以MTT比色法测定细胞活性、光学显微镜观察细胞形态变化和电子显微镜观察细胞微管等超微结构变化,观察OA所致AD样细胞效应.结果 OA可引起SH-SY5Y细胞活性下降,呈剂量和时间依赖性;光学显微镜观察SH-SY5Y细胞数量明显减少,细胞胞体皱缩,突起缩短消失;电子显微镜观察SH-SY5Y细胞微管结构破坏.结论 OA可诱导SH-SY5Y细胞建立AD样细胞模型.
