沙棘提取物对糖尿病ICR小鼠血糖及氧化功能的影响

     

摘要

目的 探讨沙棘提取物对链脲佐菌素致糖尿病(DM)小鼠血糖及氧化功能的影响.方法 腹腔一次性注射链脲佐菌素150 mg/kg制备DM小鼠模型,治疗组分别按沙棘提取物50、100、150 mg/kg每天灌胃1次,模型组和正常对照组则以等体积蒸馏水灌胃,同时观察小鼠血糖、采水量、采食量、24 h尿量.4 W后,分别测定血清和肝组织丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和一氧化氮合酶(NOS)活性.结果 与DM模型组ICR小鼠比较,沙棘提取物各组DM小鼠血糖显著降低,血清和肝组织SOD、CSH-Px、NOS活性明显提高、而MDA含量明显降低,体重下降和多饮多食症状得到改善.结论 沙棘提取物有降低血糖作用,能减轻DM小鼠肝组织氧化损伤,其机制可能与提高氧化酶活性、清除自由基、减少氧化应激有关.%Objective To detective the effects of seabucithorn procyanidins extract on the blood glucose and oxydation function in diabetic mellitus ( DM ) ICR mice induced by streptozotocin. Methods DM model was established by intraperitoneal administration of streptozotocin (STZ) at 150 mg/kg on ICR mice. Mice in treatment groups were administrated with seabucithorn procyanidins extract at 50,100,150 mg/kg respectively via gastric irrigation while the DM and normal control groups were administrated with same amount of distilled water via gastric irrigation daily. Fasting blood glucose(FBG) ,water and food intake,24 h urine output were observed and recorded. 4 weeks later,serum and hepatic malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and nitric oxide synthase (NOS) were analysed. Results Compared with DM ICR group,the FBG was reduced markedly in seabucithorn procyanidins extract groups with increased level of serum and hepatic SOD,GSH-Px and NOS while decreased MDA. Meanwhile the general condition of weight loss and polydipsia and polyphagia was improved obviously. Conclusions Seabuckthom procyanidins extract possesses the effect of reducing blood sugar in DM rats and relieving the oxidative damage of hepatic tissue. Its mechanism may associate with the effect of enhancing the activity of oxidase, cleaning free radicals and ameliorating oxidative stress.
